Palmitoylation of Plasmodium alveolins promotes cytoskeletal function. [PDF]
S-palmitoylation is a post-translational lipid modification that is widespread among Plasmodium proteins and essential for parasite development. Little is known about the contribution of palmitoylation to the function of individual parasite molecules and

Peroxiredoxin 6 mediates Gαi protein-coupled receptor inactivation by cJun kinase
Opioid receptors are important modulators of nociceptive pain. Here the authors show that opioid receptor activation recruits peroxiredoxin 6 (PRDX6) to the receptor-Gαi complex by c-Jun N-terminal kinase, resulting in Gαi depalmitoylation and enhanced ...

This study identifies palmitoylation as a novel regulatory modification of SMAD4, mediated by ZDHHC22/APT2. It activates fatty acid synthesis, creating a self‐reinforcing SMAD4–FASN–palmitate feedback loop that drives pancreatic cancer growth and enhances radiotherapy sensitivity.

Metabolic Labeling of Ras with Tritiated Palmitate to Monitor Palmitoylation and Depalmitoylation [PDF]
Metabolic labeling with tritiated palmitate is a direct method for monitoring posttranslational modification of Ras proteins with this fatty acid. Advances in intensifying screens have allowed for the easy visualization of tritium without the need for extended exposure times.

Specific oligomerization of the 5-HT1A receptor in the plasma membrane [PDF]
In the present study we analyze the oligomerization of the 5-HT1A receptor within living cells at the sub-cellular level. Using a 2-excitation Foerster Resonance Energy Transfer (FRET) method combined with spectral microscopy we are able to estimate the ...

Protein palmitoylation in cancer: molecular functions and therapeutic potential
Protein S‐palmitoylation (hereinafter referred to as protein palmitoylation) is a reversible lipid posttranslational modification catalyzed by the zinc finger DHHC‐type containing (ZDHHC) protein family.

Hepatocellular carcinoma (HCC)‐derived extracellular vesicles (EVs) enrich the metabolic enzyme ATP‐citrate lyase (ACLY). EV‐transferred ACLY enhances palmitate biosynthesis, increases the S‐palmitoylation and stability of multiple immune checkpoint proteins, augments the cellular immunosuppressive activity, and ultimately accelerates the malignant ...

Depalmitoylated Ras traffics to and from the Golgi complex via a nonvesicular pathway [PDF]
Palmitoylation is postulated to regulate Ras signaling by modulating its intracellular trafficking and membrane microenvironment. The mechanisms by which palmitoylation contributes to these events are poorly understood. Here, we show that dynamic turnover of palmitate regulates the intracellular trafficking of HRas and NRas to and from the Golgi ...

Palmitoyl Acyltransferase DHHC21 Mediates Endothelial Dysfunction in Systemic Inflammatory Response Syndrome [PDF]
Endothelial dysfunction is a hallmark of systemic inflammatory response underlying multiple organ failure. Here we report a novel function of DHHC-containing palmitoyl acyltransferases (PATs) in mediating endothelial inflammation.
