Combination treatment for glioblastoma with temozolomide, DFMO and radiation.

Journal of B.U.ON. : official journal of the Balkan Union of Oncology, 2019
Glioblastoma multiforme (GBM) is the most malignant primary brain tumor with dismal prognosis. This tumor is characterized by extensive heterogeneity, thus is difficult to treat and every established or new treatment faces significant hazard of resistance. Temozolomide (TMZ), an oral alkylating agent, is the first-line treatment for GBM, but resistance

CNS toxicity and CSF pharmacokinetics of intraventricular DFMO and MGBG in beagle dogs

Cancer Chemotherapy and Pharmacology, 1984
We have developed a beagle dog model to study the pharmacology and toxicology of anticancer drugs administered through the 3rd or lateral ventricles. A Foltz-type reservoir was implanted SC and connected by tube into a cerebral ventricle. Drugs were administered directly into the reservoir; CSF sampling of drugs administered into the ventricle was ...

Effect of difluoromethyl ornithine (DFMO) on small intestine of adult and weanling rats

Digestive Diseases and Sciences, 1987
Oral feeding of DL-difluoromethyl ornithine (DFMO) (2% in water ad libitum) for 14 days has no detectable effect on the small intestine of adult rats. Similar feeding of DFMO to weanling rat pups caused diarrhea in three to four days accompanied by a decrease in food consumption and body weight compared to age-matched controls. Significant decreases in

Effects of Starvation and Difluoromethylornithine (DFMO) on Diamine Oxidase Activity in Rat Ileum

Digestion, 2009
Starvation and difluoromethylornithine (DFMO) administration have profound affects on intestinal proliferation, ornithine decarboxylase activity, and tissue polyamine levels. Diamine oxidase activity may play a role in the regulation of proliferation, and the activity of this enzyme may be influenced by ornithine decarboxylase activity. To determine if

Concomitant DFMO and sulindac chemoprevention of colorectal adenomas: a major clinical advance

Nature Clinical Practice Oncology, 2008
In a randomized, placebo-controlled, double-blind clinical trial by Meyskens et al. the combination of difluoromethylornithine and sulindac has been shown to be strikingly effective for prevention of sporadic colorectal adenomas. This concomitant use of two drugs to suppress the progression of preneoplastic lesions represents the first major clinical ...

Chemotherapy of trypanosomiasis: the potentiation of antimonial compounds by difluoromethylornithine (DFMO).

Tropical medicine and parasitology : official organ of Deutsche Tropenmedizinische Gesellschaft and of Deutsche Gesellschaft fur Technische Zusammenarbeit (GTZ), 1991
The currently available anti-leishmanial antimonial drugs, together with other antimony compounds were individually tested, in combination with difluoromethylornithine (DFMO), against experimental murine trypanosomiasis with central nervous system involvement.

[DFMO: an alternative therapeutic agent for human African trypanosomiasis].

Bulletin de la Societe de pathologie exotique et de ses filiales, 1989
alpha-Difluoromethyl ornithine (DFMO) irreversibly inhibits ornithine decarboxylase (ODC), an essential enzyme for the synthesis of polyamines, and results in decreased biosynthesis of putrescine and spermidine, factors necessary for cellular multiplication.

Immobilization of Cell Cycle Specific Drug DFMO for Chemotherapy

1993
Cell cycle specific drugs have advantages over conventional toxic cancer drugs. Immobilization of these drugs aims at increasing their bioavailability and efficiency. DFMO (Difluoromethyl-ornithine) inhibits polyamine biosynthesis by binding to the rate limiting enzyme Ornithine decarboxcylase (ODC).
