Altered Tissue and Plasma Levels of Fibroblast Activation Protein-α (FAP) in Renal Tumours [PDF]
(1) Background: Renal cell carcinoma (RCC) is a heterogeneous and complex disease with only partial response to therapy, high incidence of metastasis and recurrences, and scarce reliable biomarkers indicative of progression and survival. Cancer-associated fibroblasts (CAFs) play an important role supporting and promoting renal cancer progression.

Fibroblast growth factor 21 (FGF21) is an important regulator of energy metabolism. FGF21 is inactivated by fibroblast activation protein (FAP). We investigated whether FGF21 and/or FAP are secreted from human white adipose tissue of individuals with ...

New Frontiers in Cancer Imaging and Therapy Based on Radiolabeled Fibroblast Activation Protein Inhibitors: A Rational Review and Current Progress [PDF]
Over the past decade, the tumor microenvironment (TME) has become a new paradigm of cancer diagnosis and therapy due to its unique biological features, mainly the interconnection between cancer and stromal cells.

This study developed a dual‐targeting biomimetic nanozyme hybrid system (Cu‐CeO2@ABs‐FTP) and integrated it with HAMA hydrogel to fabricate a microneedle patch (CAF@MN). The patch promotes fibroblast‐mediated wound repair, targets macrophages to induce M2 polarization, and targets myofibroblasts to exert anti‐fibrotic effects, thereby enabling rapid ...

Fzd7 Restrains Pink1‐Dependent Mitophagy in Suture Stem Cells to Maintain Cranial Suture Patency
How suture stem cells fail to preserve cranial suture patency remains incompletely understood. Integrated single‐cell and high‐resolution spatial transcriptomic analyses identify reduced Fzd7 expression in Prrx1+ suture stem cells as an early feature of craniosynostosis.

CAF‐derived exosomes deliver circFAD104 into TNBC cells, where it acts as a molecular scaffold that bridges the E3 ligase MARCHF8 and PGM1, promoting MARCHF8‐mediated K48‐linked ubiquitination and proteasomal degradation of PGM1. Loss of PGM1 redirects glucose‐phosphate flux from glycogen synthesis toward glycolysis, thereby driving stemness, EMT, and ...

Quantitation of fibroblast activation protein (FAP)‐specific protease activity in mouse, baboon and human fluids and organs [PDF]
The protease fibroblast activation protein (FAP) is a specific marker of activated mesenchymal cells in tumour stroma and fibrotic liver. A specific, reliable FAP enzyme assay has been lacking. FAP's unique and restricted cleavage of the post proline bond was exploited to generate a new specific substrate to quantify FAP enzyme activity. This sensitive

During pancreatitis, IL‐1α is released from necrotic acinar cells. In response to IL‐1α, pancreatic fibroblasts release chemokines and cytokines that regulate the recruitment of immune cells to the damaged organ. Furthermore, IL‐1α primes fibroblasts, resulting in increased tissue fibrosis during chronic pancreatitis.

Isolation of Fibroblast-Activation Protein-Specific Cancer-Associated Fibroblasts [PDF]
The current study is to develop a gentle and efficient method for purification of fibroblast-activation protein positive (FAP+) cancer-associated fibroblasts (CAFs) from tumor tissues. Fresh tissues were isolated from BALB/c-Nude mice bearing human liver

A novel hetero‐bivalent agent, 68Ga/177Lu‐FZ‐NF‐1, concurrently targets Nectin‐4 and FAP in triple‐negative breast cancer. It yields significant preclinical tumor regression and clinically identifies additional metastatic lesions missed by 18F‐FDG PET, demonstrating promising theranostic potential for personalized management.
