Aged ZnO Nanoparticles Promote Breast Cancer Lung Metastasis by Remodeling the Pulmonary Microenvironment Through the CXCL10/Wnt/β‐Catenin Axis

open access: yesAdvanced Science, EarlyView.
In summary, pulmonary exposure to ZnO NPs, particularly aged NPs, promotes breast cancer metastasis by remodelling the lung tumour microenvironment. Aged ZnO NPs enrich M2‐macrophages, MDSCs and activated CAFs to form an immunosuppressive metastatic niche. CXCL10 from these cells drives metastasis via Wnt/β‐catenin activation.

High Expression of FAP in Colorectal Cancer Is Associated With Angiogenesis and Immunoregulation Processes

open access: yesFrontiers in Oncology, 2020
Fibroblast activation protein α (FAP) plays an important role in tissue remodeling and helps tumor cells invade surrounding tissue. We sought to investigate FAP as a prognostic molecular marker in colorectal cancer (CRC) using immunohistochemical and ...

Long‐Term Expandable Periosteal Organoids Reconstruct a Functional Regenerative Niche That Support Hematopoietic Stem Cell Maintenance and Bone Repair

open access: yesAdvanced Science, EarlyView.
A robust periosteal organoid system enables long‐term expansion while preserving native cellular heterogeneity. These organoids recapitulate functional niche environments, support hematopoietic stem cell maintenance, and attenuate aging‐associated decline ex vivo.

Biomimetic Nanoplatform for Fibrosis Remodeling and Anti‐PD‐1 Potentiation via Cu+/Ca2+ Ion Dysregulation and TGF‐β Inhibition in PDAC

open access: yesAdvanced Science, EarlyView.
A biomimetic nanoplatform (RPm@CC‐LY) remodels fibrotic stroma via sustained TGF‐β inhibition, triggers oxidative stress and apoptosis through Cu+‐mediated Fenton chemistry and Ca2+ overload, and reprograms macrophages and dendritic cells to boost CD8+ T cell infiltration.

Histone Lactylation Drives WNK1 Expression to Promote Esophageal Squamous Cell Carcinoma Progression Through Histone H3 Ser10 Phosphorylation

open access: yesAdvanced Science, EarlyView.
In brief. Yan et al. demonstrated that the excess lactate produced by the enhanced glycolysis pathway in esophageal cancer can lactylate H3K23, H3K23la can be enriched in the WNK1 promoter region to promote WNK1 expression, thereby increasing the phosphorylation of the p‐H3 Ser10 site, and p‐H3S10 can be enriched in the AKT1 promoter region to promote ...

Increase Fibroblast Activation Protein-α, but not Lowering of FGF Receptor and/or Co-Receptor, Causes Increase in Fibroblast Growth Factor-21 in T2D Independent on Diabetes Duration [PDF]

open access: yesAin Shams Medical Journal
Background: Diabetes mellitus is a major global public health issue. T2DM patients are associated with increased Fibroblast growth Factor 21 (FGF21).

Single‐Nucleus RNA Sequencing Identifies CREM as a Brake on Myofiber Hypertrophy in Chickens

open access: yesAdvanced Science, EarlyView.
Single‐nucleus RNA sequencing on 91 144 nuclei from postnatal chicken pectoralis major between fast‐growing broilers (FG) and slow‐growing layers (SG) identifies the transcriptional factor CREM as a novel brake of myofiber hypertrophy. Mechanistically, functional validation experiments confirm that CREM activates the promoter of JUN and CREM deletion ...

Phenotypic Heterogeneity of Cancer Associated Fibroblasts in Cervical Cancer Progression: FAP as a Central Activation Marker

open access: yesCells
Cervical cancer (CC) is the fourth leading cancer among women and is one of the principal gynecological malignancies. In the tumor microenvironment, cancer-associated fibroblasts (CAFs) play a crucial role during malignant progression, exhibiting a ...

A model‐based approach leveraging in vitro data to support dose selection from the outset: A framework for bispecific antibodies in immuno‐oncology

open access: yesCPT: Pharmacometrics & Systems Pharmacology, 2023
FAP‐4‐1BBL is a bispecific antibody exerting 4‐1BB‐associated T‐cell activation only while simultaneously bound to the fibroblast activation protein (FAP) receptor, expressed on the surface of cancer‐associated fibroblasts.

A specific, transmembrane interface regulates fibroblast activation protein (FAP) homodimerization, trafficking and exopeptidase activity

open access: yesBiochimica et Biophysica Acta (BBA) - Biomembranes, 2016
Fibroblast activation protein (FAP) is a cell-surface serine protease which promotes invasiveness of certain epithelial cancers and is therefore a potential target for cancer drug development and delivery. Unlike dipeptidyl peptidase IV (DPPIV), FAP exhibits prolyl endopeptidase activity and is active as a homodimer with specificity for type I collagen.
