This work examines the interaction between farnesyltransferase (FTase) and cell‐permeable CaaX peptides. Particularly, peptide CaaX‐1 is presented as efficient binder to FTase capable to inhibit further interaction of other CaaX motif–bearing peptides. Moreover, biological effects require an intact CaaX motif.

Supersulfides: A Promising Therapeutic Approach for Autoinflammatory Diseases
ABSTRACT Supersulfides are molecular species characterized by catenated sulfur moieties, including low‐molecular‐weight and protein‐bound supersulfides. Emerging evidence suggests that these molecules, abundantly present in diverse organisms, play essential roles far beyond their chemical properties, such as functions in energy metabolism, protein ...

Nitrogenous bisphosphonate inhibitors of farnesyl disphosphate synthase have been used clinically for treatment of bone disease. Because many of their effects may be mediated by depletion of geranylgeranyl diphosphate, our group has sought compounds ...

Geranylgeranylation but not GTP loading determines rho migratory function in T cells
Rho GTPases orchestrate signaling pathways leading to cell migration. Their function depends on GTP loading and isoprenylation by geranylgeranyl pyrophosphate (GGpp).

Lovastatin, a competitive inhibitor of 3-hydroxy-3- methylglutaryl (HMG)- CoA reductase, inhibits the conversion of mevalonate from HMG-CoA. Previously, we have reported that lovastatin treatment induced the occurrence of apoptosis and differentiation in

Background: Available in vitro and in vivo methods for verifying protein substrates for posttranslational modifications via farnesylation or geranylgeranylation (for example, autoradiography with 3H-labeled anchor precursors) are time consuming (weeks ...

Geranylgeranylation but not GTP-loading of Rho GTPases determines T cell function
Rho guanosine triphosphatases (GTPases) orchestrate signaling pathways leading to cell migration. They are typically responsible for the organization of actin filaments that support actomyosin contractility and cell-body translocation.

The activity of small GTP-binding proteins is regulated by a critical step in posttranslational processing, namely, the addition of isoprenoid lipids farnesyl and geranylgeranyl, mediated by the enzymes farnesyltransferase (FTase) and ...
