Background: Aberrant O-glycosylation of IgA1 plays an important role in IgA nephropathy pathogenesis. Previous proteomic studies analyzed O-glycans of the circulating IgA1 hinge region and found that the N-acetylgalactosamine (GalNAc) and galactose ...

The Critical Role of GALNTs‐Regulated O‐GalNAc Glycosylation in Cancer Malignancy
The GALNT family initiates mucin‐type O‐GalNAc glycosylation in the Golgi apparatus and how regulatory mechanisms, including GALNT relocalization and microRNA control, modulate this process. Dysregulated O‐GalNAc glycosylation alters key cellular behaviors and promotes cancer malignancy, highlighting its relevance to tumor progression as well as its ...

Glycoproteomics-Based Identification of Cancer Biomarkers [PDF]
Protein glycosylation is one of the most common posttranslational modifications in mammalian cells. It is involved in many biological pathways and molecular functions and is well suited for proteomics-based disease investigations. Aberrant protein glycosylation may be associated with disease processes.

Primary liver cancer, mainly comprising hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (ICC), remains a major global health problem. Although ICC is clinically different from HCC, their molecular differences are still largely unclear.

Inhaled Angiopoietin‐Like 4 Antisense Oligonucleotide Therapy for Lung Injury and Fibrosis
Inhaled antisense oligonucleotides targeting ANGPTL4 attenuate inflammation, preserve barrier integrity, and reduce fibrosis in both infectious and sterile lung injury models. Integrated transcriptomic analysis reveals a conserved ANGPTL4‐regulated gene network coordinating hypoxic, inflammatory, apoptotic, and stress‐response pathways.

Quantitative Serum Glycomics of Esophageal Adenocarcinoma, and Other Esophageal Disease Onsets [PDF]
Aberrant glycosylation has been implicated in various types of cancers and changes in glycosylation may be associated with signaling pathways during malignant transformation.

This study identifies B4GALNT4 as a key driver of prostate cancer (PCa). It shows that B4GALNT4 glycosylates PDK1 protein at the N531 site, which stabilizes the PDK1 and constitutively activates the PI3K‐AKT pathway. This mechanism promotes tumor cell proliferation, migration, and invasion. The findings establish the B4GALNT4‐PDK1 glycosylation axis as

Engineering Orthogonal Polypeptide GalNAc-Transferase and UDP-Sugar Pairs [PDF]
O-Linked α-N-acetylgalactosamine (O-GalNAc) glycans constitute a major part of the human glycome. They are difficult to study because of the complex interplay of 20 distinct glycosyltransferase isoenzymes that initiate this form of glycosylation, the ...

Improved strategy for large scale isolation of sialylglycopeptide (SGP) from egg yolk powder [PDF]
Chicken egg yolk is an easily available source for the isolation of sialylglycopeptides (SGP) carrying homogenous biantennary N-glycans. This approach has gained much attention in the last decade since these SGPs can easily be used for the semi-synthesis

Pushing Frontiers for Proteoglycans
ABSTRACT Almost half of proteins have one or more post‐translational glycosylation modifications involving the addition of glycan structures. Proteoglycans are a class of glycosylated proteins with one or more glycosaminoglycan (GAG) chains covalently attached to the core protein.
