Apoptotic and anti-proliferative effect of guanosine and guanosine derivatives in HuT-78 T lymphoma cells

open access: yes, 2020
The effects of 100 μM of 3',5'-cGMP, cAMP, cCMP, and cUMP as well as of the corresponding membrane-permeant acetoxymethyl esters on anti-CD3-antibody (OKT3)-induced IL-2 production of HuT-78 cutaneous T cell lymphoma (Sézary lymphoma) cells were analyzed.

Comparison of the stabilities and solution structures of metal ion complexes formed with 5'-di- and 5'-triphosphates of purine nucleosides [PDF]

open access: yes, 2003
Nucleotides and metal ions are involved in the basic metabolic processes of life, participating as substrates in enzymatic reactions in the form of metal ion complexes.

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Nucleoside diphosphate kinases fuel dynamin superfamily proteins with GTP for membrane remodeling [PDF]

open access: yes, 2014
International audienceDynamin superfamily molecular motors use guanosine triphosphate (GTP) as a source of energy for membrane-remodeling events. We found that knockdown of nucleoside diphosphate kinases (NDPKs) NM23-H1/H2, which produce GTP through ...

UHPLC–MS based metabolomics study of the ErZhi formula on skeletal muscle against osteosarcopenia focusing on energy metabolism

open access: yesAnimal Models and Experimental Medicine, EarlyView.
ErZhi formula (EZF) is composed of Ligustri Lucidi Fructus (LLF) and Ecliptae herba (EH). Ovariectomized (OVX) and sham rats received 12‐week intragastric treatment with EZF, alendronate, and vehicle saline. Gastrocnemius tissues were harvested to systematically evaluate pharmacodynamic changes in skeletal muscle morphology, mitochondrial ...

A Modular Nucleoside Kinase Cascade for the Synthesis of Ribonucleoside Triphosphates

open access: yesBiotechnology and Bioengineering, EarlyView.
Damm et al. report standardized enzymatic cascades for production of all canonical nucleoside triphosphates from the corresponding nucleosides, using kinase‐catalyzed phosphorylation from acetyl phosphate. Reactions afforded ∼ 100 mM product in near‐quantitative conversion.

Membrane‐Permeable Nucleoside T‐1106 Diphosphate and Triphosphate Analogues as Antiviral Pronucleotides

open access: yesChemistry – A European Journal, EarlyView.
Favipiravir (T‐705) and the non‐fluorinated counterpart (T‐1106) are antiviral agents that inhibit the RNA‐dependent RNA polymerase (RdRp) of various RNA viruses. The antiviral efficacy of nucleoside analogues is strongly dependent on their intracellular activation by cellular kinases to produce their corresponding triphosphate metabolites (T‐705‐RTP ...

Discontinuing Long‐Term Denosumab in Treating Fragile Bone: Why, for Whom, and How?

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
Discontinuation of denosumab (Dmab) may be necessary due to adverse events or an unfavorable long‐term risk–benefit profile. However, accumulating evidence demonstrates pronounced rebound phenomena after withdrawal, most notably a marked increase in multiple vertebral fractures, and, in some reports, elevated mortality.

Oncogenic KRAS Rewires Stress Granule Dynamics: Mechanisms and Therapeutic Opportunities

open access: yesThe Kaohsiung Journal of Medical Sciences, EarlyView.
ABSTRACT Stress granules (SGs) are dynamic, membrane‐less structures that form in response to various cellular stresses, including metabolic, oxidative, and therapeutic challenges. They function as adaptive hubs and reorganize protein synthesis and signaling networks to help cells survive under stress. In cancer, these condensates are often hijacked to
