DADA Enhances CD8+ T Cell Stemness to Improve Anti‐Tumor Immunity and Immunotherapy Efficacy

open access: yesAdvanced Science, EarlyView.
Diisopropylamine dichloroacetate (DADA) enhances CD8+ T cell stemness by improving OXPHOS and mitochondrial fitness in a PDK1‐depenpendent manner. This metabolic shift strengthens CD8+ T cell anti‐tumor immunity, improves responses to PD‐1 blockade, and endows CAR‐T cells with prolonged persistence and resistance to terminal exhaustion, highlighting a ...

Effects of Salicylic Acid on the Metabolism of Mitochondrial Reactive Oxygen Species in Plants

open access: yesBiomolecules, 2020
Different abiotic and biotic stresses lead to the production and accumulation of reactive oxygen species (ROS) in various cell organelles such as in mitochondria, resulting in oxidative stress, inducing defense responses or programmed cell death (PCD) in

GENETICS OF YEAST HEXOKINASE [PDF]

open access: yesGenetics, 1977
ABSTRACT Two independent isolates of Saccharomyces cerevisiae lacking hexokinase activity (EC 2.7.1.1) are described. Both mutant strains grow on glucose but are unable to grow on fructose, and contain two mutant genes h×k1 and h×k2 each. The mutations are recessive and noncomplementing.

AMP-activated protein kinase (AMPK) action in skeletal muscle via direct phosphorylation of PGC-1α [PDF]

open access: yes, 2007
Activation of AMP-activated kinase (AMPK) in skeletal muscle increases glucose uptake, fatty acid oxidation, and mitochondrial biogenesis by increasing gene expression in these pathways.

Leveraging Macrophage Metabolic Reprogramming for Enhanced Anti‐Tumor Immunity

open access: yesAdvanced Science, EarlyView.
Tumor‐associated macrophages (TAMs) are key regulators of the tumor microenvironment (TME), with their metabolic states playing a critical role in tumor progression or regression. This review summarizes current understanding of TAM metabolic plasticity alongside cutting‐edge bioengineering innovations, outlining a roadmap for transforming the ...

Bifunctional Artificial Enzymes‐Loaded Microgels With LOX‐ and CAT‐Like Activities for Metabolic Reprogramming and Scarless Wound Repair

open access: yesAdvanced Science, EarlyView.
A bifunctional lactate oxidase‐like and catalase‐like artificial enzyme (Metazyme) is integrated into a rod‐shaped microgel (MetaRgel) to enable cascade lactate oxidation and oxygen regeneration. By reprogramming the wound metabolic microenvironment, MetaRgel alleviates excessive lactate accumulation, oxidative stress, hypoxia, and inflammation ...

Exploring Non-Metabolic Functions of Glycolytic Enzymes in Immunity

open access: yesFrontiers in Immunology, 2017
At the beginning of the twentieth century, discoveries in cancer research began to elucidate the idiosyncratic metabolic proclivities of tumor cells (1).

Activators of Yeast Hexokinase

open access: yesJournal of Biological Chemistry, 1971
Abstract The rate of the ATP:glucose transphosphorylase reaction catalyzed by yeast hexokinase, isozyme Pii, is inhibited by lowering the pH of the reaction below 7.0, especially at suboptimal concentrations of ATP. This effect of acidity is largely overcome by activators such as orthophosphate, citrate, malate, 3-phosphoglycerate, and riboside ...

Metabolic Imbalance Triggers Adaptive Remodeling to Accelerate Diploidization in Murine Haploid Embryonic Stem Cells

open access: yesAdvanced Science, EarlyView.
In this article, Shuai and colleagues demonstrate that metabolic remodeling drives self‐diploidization in murine haploid ESCs (haESCs). Mitochondrial dysfunction and imbalanced pyruvate metabolism underlie this process. Genome‐wide screening using haESCs identifies key mitochondrial quality‐control related genes, enabling a metabolism‐based medium that

Mitochondrial Enzymes Mimetic Ultrasmall Palladium Nanozymes Prevent Senescence and Neurodegeneration Through Metabolic Reprogramming

open access: yesAdvanced Science, EarlyView.
Simply constructed polyvinylpyrrolidone (PVP) modified palladium nanoparticles (PdP NPs) as cytochrome c oxidase (CcO) and superoxide dismutase (SOD) mimic are promising multifunctional nanoreactors to boost mitochondrial respiration chain function and refine homeostasis via regulation of mitochondrial dynamics and proteostasis sensor UPRmt, which ...
