Lenvatinib resistance poses a major challenge in advanced hepatocellular carcinoma (HCC). This study reveals that DNMT2 upregulation is a key driver, which stabilizes ACLY mRNA via m5C modification and activates the Notch signaling pathway. Crucially, combining ACLY inhibitors with lenvatinib overcomes resistance and suppresses tumors, offering a ...

Plant responses to abiotic stress regulated by histone acetylation
In eukaryotes, histone acetylation and deacetylation play an important role in the regulation of gene expression. Histone acetylation levels are reversibly regulated by histone acetyltransferases (HATs) and histone deacetylases (HDACs).

Lysine acetyltransferase Gcn5-B regulates the expression of crucial genes in Toxoplasma and its function is regulated through lysine acetylation [PDF]
Indiana University-Purdue University Indianapolis (IUPUI)Histone acetylation has been linked to developmental changes in gene expression and is a validated drug target of apicomplexan parasites, but little is known about the roles of individual histone ...

Intervertebral disc degeneration is triggered by ACSL4 accumulation‐mediated ferroptosis of nucleus pulposus cells due to CMA dysfunction. KAT2B promotes ACSL4 degradation via CMA through acetylation. AAV‐mediated LAMP2A delivery or engineered exosomes rescue nucleus pulposus cell senescence and disc degeneration.

Histone posttranslational modifications (PTMs) help regulate DNA templated processes; however, relatively little work has unbiasedly explored the single-molecule combinations of histone PTMs, their dynamics on short timescales, or how these preexisting ...

MYST-family histone acetyltransferases: beyond chromatin
Covalently modifying a protein has proven to be a powerful mechanism of functional regulation. N-epsilon acetylation of lysine residues was initially discovered on histones and has been studied extensively in the context of chromatin and DNA metabolism, such as transcription, replication and repair.

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome

Epigenomic regulation of heart failure: integrating histone marks, long noncoding RNAs, and chromatin architecture. [PDF]
Epigenetic processes are known to have powerful roles in organ development across biology. It has recently been found that some of the chromatin modulatory machinery essential for proper development plays a previously unappreciated role in the ...

Phase I dose-escalation study of the mTOR inhibitor sirolimus and the HDAC inhibitor vorinostat in patients with advanced malignancy. [PDF]
Preclinical models suggest that histone deacetylase (HDAC) and mammalian target of rapamycin (mTOR) inhibitors have synergistic anticancer activity. We designed a phase I study to determine the safety, maximum tolerated dose (MTD), recommended phase II ...

This study demonstrates that somatic PIK3CA mutations suppress PPT1 expression via activation of the PI3K–AKT–c‐JUN axis. This reduction in PPT1 weakens its interaction with P300, thereby increasing palmitoylation at C1176 of P300 and protecting P300 from lysosomal degradation.
