ARID5A stabilizes Indoleamine 2,3-dioxygenase expression and enhances CAR T cell exhaustion in colorectal cancer

open access: yesTranslational Oncology
Resistance to chimeric antigen receptor (CAR) T-cell therapy remains a significant challenge in the treatment of solid tumors. This resistance is attributed to various factors, including antigen loss, immunosuppressive tumor microenvironment, and ...

Traditional Chinese Medicine for lung cancer: Mechanisms, clinical evidence, and future perspectives

open access: yesPrecision Medical Sciences, Volume 15, Issue 3, Page 206-226, September 2026.
Graphical abstract represents the risk factors for lung cancer, TCM efficacy and clinical outcomes to future challenges. Abstract Lung cancer remains a leading cause of global cancer mortality. Despite advances in conventional treatments such as surgery, chemotherapy, targeted therapy, and immunotherapy, challenges including drug resistance, toxicity ...

Resolving Endoplasmic Reticulum‐Protein Misfolding Restores Corticosteroid Sensitivity in Experimental Models of Severe Asthma

open access: yesAllergy, Volume 81, Issue 9, Page 3221-3235, September 2026.
This study investigates the role of ER stress in steroid resistance in severe asthma. Results from advanced 3D models of human bronchial epithelial cells, steroid‐resistant severe asthma mouse models, and human severe asthma sputum samples show that treatment with ICS in combination with the ER stress inhibitor 4‐PBA significantly enhances steroid ...

The Airway Transcriptome in Type 2 Cytokine Biomarker‐High and ‐Low Severe Asthma

open access: yesAllergy, Volume 81, Issue 9, Page 3300-3317, September 2026.
This study distinguishes severe asthma–related airway transcriptomic changes from the effects of high‐dose ICS. Most dysregulated genes in severe asthma show concordance with corticosteroid responses. Corticosteroid‐resistant T2‐high asthma featured T2‐high and epithelial barrier gene upregulation, while T2‐low asthma featured Th1/IL‐17 signatures. ASM,

GCN2 in proteostasis: structural logic, signalling networks and disease

open access: yesThe FEBS Journal, Volume 293, Issue 18, Page 5627-5644, September 2026.
Threats to protein synthesis activate the kinase GCN2, initiating the integrated stress response (ISR). GCN2 is triggered by stalled ribosomes and uncharged tRNAs, which accumulate when amino acids are scarce. The ISR adjusts cellular physiology by promoting redox balance, protein quality control, and mitochondrial optimisation.

Oxetane Promise Delivered: Discovery of Long-Acting IDO1 Inhibitors Suitable for Q3W Oral or Parenteral Dosing

open access: yes
3,3-Disubstituted oxetanes have been utilized as bioisosteres for gem-dimethyl and cyclobutane functionalities. We report the discovery of a novel class of oxetane indole-amine 2,3-dioxygenase (IDO1) inhibitors suitable for Q3W (once every 3 weeks) oral ...
