Druggable Sterol Metabolizing Enzymes in Infectious Diseases: Cell Targets to Therapeutic Leads

open access: yesBiomolecules
Sterol biosynthesis via the mevalonate-isoprenoid pathway produces ergosterol (24β-methyl cholesta-5,7-dienol) necessary for growth in a wide-range of eukaryotic pathogenic organisms in eukaryotes, including the fungi, trypanosomes and amoebae, while ...

Steroidal antibiotics are antimetabolites of Acanthamoeba steroidogenesis with phylogenetic implications

open access: yesJournal of Lipid Research, 2019
Pathogenic organisms may be sensitive to inhibitors of sterol biosynthesis, which carry antimetabolite properties, through manipulation of the key enzyme, sterol methyltransferase (SMT).

Sterol Metabolism Differentially Contributes to Maintenance and Exit of Quiescence

open access: yesFrontiers in Cell and Developmental Biology, 2022
Nutrient starvation initiates cell cycle exit and entry into quiescence, a reversible, non-proliferative state characterized by stress tolerance, longevity and large-scale remodeling of subcellular structures.

Microbial Test-System for Screening of Inhibitors of Sterol Biosynthesis

open access: yesАнтибиотики и Химиотерапия, 2020
New effective economic microbial test-system for screening of microbial metabolites, that are inhibitors of sterol biosynthesis, is proposed. It is based on cultivation of fungal strain Acremonium fusidioides (former Fusidium coccineum), that produces ...

Functional analysis of two sterol regulatory element binding proteins in Penicillium digitatum. [PDF]

open access: yesPLoS ONE, 2017
The sterol regulatory element binding proteins (SREBPs) are key regulators for sterol homeostasis in most fungi. In the citrus postharvest pathogen Penicillium digitatum, the SREBP homolog is required for fungicide resistance and regulation of CYP51 ...

Plant Growth Retardants as Inhibitors of Sterol Biosynthesis in Tobacco Seedlings [PDF]

open access: yesPlant Physiology, 1974
Three plant-growth retardants 2'-isopropy1-4'-(trimethylammonium chloride)-5-methylphenylpiperidine carboxylate (Amo 1618), beta-chloroethyltrimethylammonium chloride, and tributyl-2, 4-dichlorobenzylphosphonium chloride were tested for their effects on sterol production in, and growth of tobacco (Nicotiana tabacum) seedlings.

Engineering Nanoplatforms for Alzheimer's Disease Detection via Biomolecular Corona Proteomic and Lipidomic Profiling

open access: yesAdvanced Healthcare Materials, EarlyView.
Engineered nanoparticles capture disease‐specific biomolecular coronas that uncover hidden molecular features of Alzheimer's disease. Combined proteomic and lipidomic analyses reveal a characteristic shift in ribosomal machinery and energy metabolism, generating a multiomic fingerprint that supports accurate disease detection and opens new ...

Targeting Ergosterol biosynthesis in Leishmania donovani: essentiality of sterol 14 alpha-demethylase. [PDF]

open access: yesPLoS Neglected Tropical Diseases, 2015
Leishmania protozoan parasites (Trypanosomatidae family) are the causative agents of cutaneous, mucocutaneous and visceral leishmaniasis worldwide. While these diseases are associated with significant morbidity and mortality, there are few adequate ...

Targeting the HSPA8‐CMA‐ATP6V1A Axis Triggers Lysosomal Hyperacidification and Catastrophic Vacuolation in Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Our experimental evidence supports a model in which ALO targets the HSPA8‐CMA‐ATP6V1A axis to induce lysosomal hyperacidification and initiate osmotic and lipidomic stress. These changes are associated with LMP and loss of lysosomal integrity in prostate cancer cells.
