The mitochondrial intermembrane space – a permanently proteostasis-challenged compartment

open access: yesBiological Chemistry
Abstract The mitochondrial intermembrane space (IMS) houses proteins essential for redox regulation, protein import, signaling, and energy metabolism. Protein import into the IMS is mediated by dedicated pathways, including the disulfide relay pathway for oxidative folding.

Single‐Nucleus RNA Sequencing Identifies CREM as a Brake on Myofiber Hypertrophy in Chickens

open access: yesAdvanced Science, EarlyView.
Single‐nucleus RNA sequencing on 91 144 nuclei from postnatal chicken pectoralis major between fast‐growing broilers (FG) and slow‐growing layers (SG) identifies the transcriptional factor CREM as a novel brake of myofiber hypertrophy. Mechanistically, functional validation experiments confirm that CREM activates the promoter of JUN and CREM deletion ...

Age-related immune states and PD-1 blockade: mechanisms and strategies for the elderly

open access: yesJournal for ImmunoTherapy of Cancer
Aging impairs antitumor immunity and may reduce the efficacy of immune checkpoint inhibitors (ICIs). However, the underlying mechanisms remain unclear. Building on our recent findings, we review three key mechanisms of CD8+ T-cell aging: elevated T-cell ...

Lactylation of HSC70 Impairs Chaperone‐Mediated Autophagy to Promote Pyrin Inflammasome‐Dependent PANoptosis in Macrophages During Sepsis

open access: yesAdvanced Science, EarlyView.
In sepsis, lactate accumulation drives HSC70 K561 lactylation, which impairs chaperone‐mediated autophagy and blocks CUX1 degradation. Accumulated CUX1 transcriptionally upregulates Pyrin, triggering PANoptosis in macrophages and exacerbating inflammatory storm and organ injury.

Modulation of the activity of human mitochondrial protease complex ClpXP as potential therapeutic strategy for cancer

open access: yes, 2023
Mitochondrial chaperones and proteases, crucial for mitochondrial proteostasis, are overexpressed in most tumor types, and are involved in cell metabolic reprogramming that allows evasion of apoptosis and increased survival (1).

Animal models for investigating post‐stroke cognitive impairment: A neurovascular trafficking perspective

open access: yesAnimal Models and Experimental Medicine, EarlyView.
Animal models enable mechanistic testing of how ischemic injury disrupts neurovascular trafficking and drives cognitive vulnerability after stroke. Complementary models capture distinct vascular, cellular, white matter, and cognitive features of PSCI.

Secretopathies emerge as a new class of neurocristopathies

open access: yesDevelopmental Dynamics, EarlyView.
Abstract Neural crest cells are a transient embryonic population of cells that give rise to a wide range of structures, including craniofacial cartilage and bone, peripheral neurons and glia, as well as components of the cardiac outflow tract, among others.

Redox regulation of GRPEL2 nucleotide exchange factor for mitochondrial HSP70 chaperone

open access: yesRedox Biology, 2018
Mitochondria are central organelles to cellular metabolism. Their function relies largely on nuclear-encoded proteins that must be imported from the cytosol, and thus the protein import pathways are important for the maintenance of mitochondrial ...

Mitochondrial Translation Efficiency Controls Cytoplasmic Protein Homeostasis

open access: yes, 2018
Cellular proteostasis ismaintained via the coordinated synthesis, maintenance, and breakdown of proteins in the cytosol and organelles. While biogenesis of the mitochondrial membrane complexes that execute oxidative phosphorylation depends on cytoplasmic

A shining starlet: Nematostella vectensis as a model for developmental, regenerative, and comparative biology

open access: yesDevelopmental Dynamics, EarlyView.
Abstract The sea anemone, Nematostella vectensis, has been used as a model organism in developmental biology studies for many years. This estuarine species has the notable capacity to regenerate its full body plan from small pieces throughout life. Nematostella have been described as having a great degree of cellular plasticity.
