Single‐Cell Transcriptomic Identification of Drug‐Tolerant Persister Mechanisms in Capecitabine‐Treated Triple‐Negative Breast Cancer

open access: yesComputational and Systems Oncology, Volume 6, Issue 1, December 2026.
ABSTRACT Triple‐negative breast cancer (TNBC) is an aggressive subtype defined by the absence of estrogen receptor (ER), progesterone receptor (PR), and HER2 expression. This lack of receptors limits targeted therapies and contributes to high rates of recurrence.

Integration of Chemical Probes and Degradomics Reveals That Cathepsin B Functions Primarily as a Dipeptidyl Carboxypeptidase in Macrophages

open access: yesChemMedChem, Volume 21, Issue 21, 12 November 2026.
An integrated chemical biology and degradomics approach reveals the proteolytic landscape of cathepsin B in mouse macrophages. Z‐RK‐AOMK perturbs the cysteine cathepsin network and reveals that cathepsin B predominantly exhibits dipeptidyl carboxypeptidase activity in cells.

Piezo1‐mediated unfolded protein response in goblet cells regulates intestinal mucus barrier homeostasis through the Ca2+–IRE1α–XBP1 pathway

open access: yesClinical and Translational Medicine, Volume 16, Issue 11, November 2026.
This study demonstrates that activation of Piezo1 channels in goblet cells by intestinal mechanical stimulation modulates ER calcium homeostasis via calcium ion (Ca2+) flux, which in turn regulates Mucin2 production and synthesis through the IRE1α‐XBP1. Intraperitoneal administration of Yoda1 alleviates mucus barrier impairment in DSS/WAS‐treated mice.

Dichloroacetate Improves Animal Survival, Growth, Neuromuscular Activity, Mitochondrial Stress and Physiology, and Elevated Lactate in C. elegans pdha‐1 and dld‐1 RNAi Models of Pyruvate Dehydrogenase Complex Deficiency (PDCD)

open access: yesJournal of Inherited Metabolic Disease, Volume 49, Issue 6, November 2026.
ABSTRACT Pyruvate dehydrogenase complex (PDHc) deficiency (PDCD) is a primary mitochondrial disorder characterized by neurodevelopmental disability, altered intermediary metabolism, and early mortality. Dichloroacetate (DCA), a pyruvate analog, activates PDHc and remains under clinical investigation for treatment of PDCD.

The Role of Astrocytes in the Inflammatory Response in Neurodiseases

open access: yesMedComm, Volume 7, Issue 11, November 2026.
Reactive astrocyte‐mediated pathogenic mechanisms in neurodegenerative diseases Schematic representation of key astrocytic pathways contributing to pathology in the pathogenesis of AD, PD, ALS, and HD. In AD, astrocytes exhibit Aβ plaque‐associated activation characterized by GFAP/S100β upregulation, accompanied by impaired EAAT2‐mediated glutamate ...

Conserved IQ motifs, divergent apo‐calmodulin recognition in UBE3B and UBE3C

open access: yesProtein Science, Volume 35, Issue 11, November 2026.
Abstract Calmodulin (CaM) regulates diverse signaling proteins through recognition of short linear motifs, including isoleucine‐glutamine (IQ) motifs that commonly bind Ca2+‐free CaM (apo‐CaM). The homologous to E6AP carboxyl terminus (HECT) E3 ubiquitin ligases ubiquitin protein ligase E3B (UBE3B) and ubiquitin protein ligase E3C (UBE3C) are closely ...
