TGF‐β Inhibitor Potentiates Osimertinib‐Induced Anti‐Tumor Immunity in Egfr‐Mutant Lung Cancer

open access: yesCancer Science, Volume 117, Issue 5, Page 1260-1272, May 2026.
Osimertinib enhances CD8+ T cell activity but also upregulates immunosuppressive TGF‐β in Egfr‐mutant lung tumors. This study demonstrates that TGF‐β inhibition with nintedanib or vactosertib potentiates osimertinib‐induced CD8+ T cell–mediated immunity and reduce CD206+ macrophages. These findings provide a strong rationale for novel immunotherapeutic

Targeting Inflammation and Immune Regulation in Chronic Inflammation Associated Cancers

open access: yesCancer Science, Volume 117, Issue 5, Page 1209-1222, May 2026.
Chronic inflammation sustains activation of key inflammatory pathways, including Nuclear factor kappa B (NF‐κB), Interleukin‐6‐signal transducer and activator of transcription 3 (IL6‐STAT3), Phosphoinositide 3‐kinase/Akt (PI3K/Akt), and Wingless/β‐catenin (Wnt/β‐catenin) signaling. This promotes immune dysregulation through the expansion and functional

Clinical Pharmacology and Translational Science Considerations in the Development of Dual‐Payload Antibody Drug Conjugates

open access: yesClinical and Translational Science, Volume 19, Issue 5, May 2026.
ABSTRACT Antibody‐drug conjugates (ADCs) have evolved in the last decade or two from carrying a single chemotherapy payload to carrying highly potent tubulin inhibitors and DNA damaging agents. While the latter have shown success in the clinic, efficacy has been constrained by therapeutic resistance, limited tumor penetration, and suboptimal immune ...

Molecular Mechanism of Caspase‐8–Dependent Interleukin‐18 Activation in Pancreatic Cancer Cells Induced by 5‐Fluorouracil and Nutrient Starvation

open access: yesGenes to Cells, Volume 31, Issue 3, May 2026.
Nutrient starvation triggers pyroptosis with cleavage of pro‐IL‐18 and GSDMD in pancreatic cancer cells, and treatment of 5‐FU enhances this process. As a result, active IL‐18 is released and leads to inflammation. ABSTRACT Interleukin‐18 (IL‐18) is a pro‐inflammatory cytokine, and higher IL‐18 expression in pancreatic tumors is associated with poor ...

Defining the tumor microenvironment of non‐small cell lung cancer

open access: yesImmunology &Cell Biology, Volume 104, Issue 5, Page 499-518, May‐June 2026.
The tumor microenvironment (TME) in non‐small cell lung cancer is highly heterogeneous, both at a patient level and tumor microenvironment resolution. Defining the tumor microenvironment from patient‐responders and non‐responders will aid in the development of targeted therapeutics specific for each individual patient.
