This study presents a novel 4D bioprinting platform for engineering biomimetic musculoskeletal grafts. By tuning the mechanical properties of support baths, we enhance tissue fusion, collagen alignment, and cell differentiation. Using this strategy, we successfully fabricate scaled‐up, anisotropic tissues such as meniscus, articular cartilage, and ...

HMGB2–RAD21 Axis Promotes Fibro/Adipogenic Progenitor Proliferation and Regulates Fat Infiltration
This study constructed the first developmental atlas of embryonic skeletal muscle fibro/adipogenic progenitors (FAPs) and identified an HMGB2+ FAPs subpopulation that regulates FAP pool size and muscle homeostasis. HMGB2 directly targets the RAD21 promoter, and its knockout significantly reduces FAP numbers, thereby lowering the potential for ...

Arsenic Inhibits Myogenic Differentiation and Muscle Regeneration
The incidence of low birth weights is increased in offspring of women who are exposed to high concentrations of arsenic in drinking water compared with other women. We hypothesized that effects of arsenic on birth weight may be related to effects on myogenic differentiation. We investigated the effects of arsenic trioxide (As2O3) on the myogenic ...

Muscle-derived TRAIL negatively regulates myogenic differentiation
TNF-related apoptosis-inducing ligand (TRAIL) is known to induce apoptosis in cancer cells, although non-apoptotic functions have also been reported for this cytokine in various cell types. TRAIL and its receptor TRAIL-R2 are expressed in skeletal muscles, but a potential role of muscle-derived TRAIL in myogenesis has not been explored.

A muscle‐bone endocrine pathway in aging is revealed in which extracellular vesicles released from atrophic skeletal muscle (Aged‐SKM‐EVs) inhibit bone formation. These EVs deliver miR‐125a‐5p to osteoblasts, thereby suppressing the SIRT7‐Sp7 signaling axis and osteogenic differentiation.

Comparative Single‐Cell Transcriptomic Atlas Reveals the Genetic Regulation of Reproductive Traits
A cross‐species single‐cell transcriptomic atlas of reproductive and central nervous system tissues from sheep and humans reveals conserved cellular programs and regulatory networks that regulated fertility. Integration with GWAS for sheep lifetime average litter size identifies UNC5–SLIT–BMP signaling as a core pathway coordinating neuroendocrine ...

Dynamic remodeling of septin structures fine-tunes myogenic differentiation
Summary: Controlled myogenic differentiation is integral to the development, maintenance and repair of skeletal muscle, necessitating precise regulation of myogenic progenitors and resident stem cells.

To determine the role of mutant SOD1 gene (SOD1G93A) on muscle cell differentiation, we derived C2C12 muscle cell lines carrying a stably transfected SOD1G93A gene under the control of a myosin light chain (MLC) promoter-enhancer cassette.

Slc2a6 regulates myoblast differentiation by targeting LDHB
Background Type 2 diabetes mellitus is a global health problem. It often leads to a decline in the differentiation capacity of myoblasts and progressive loss of muscle mass, which in turn results in deterioration of skeletal muscle function.

Distinct Immunomodulatory Strategies Guide Mesenchymal Stromal/Stem Cell‐Mediated Bone Regeneration
Bone regeneration by mesenchymal stem cells is strongly influenced by immune signals. This study shows that priming stem cells with regulatory immune cells or an inflammation‐resolving lipid molecule enhances bone formation through distinct immune pathways.
