Method for isolation of 2′-O-methylribonucleosides and N1-methyladenosine from ribonucleic acid

Abstract N1-methyladenosine and the 2′-O-methyl derivatives of adenosine, uridine, guanosine, and cytidine have been isolated from yeast soluble RNA. A nucleoside mixture obtained by enzymic hydrolysis of the RNA was resolved by means of chromatography on partition columns into major fractions.

We describe the selective analysis of N-methylated ribonucleosides, i.e., N1-methyl adenosine (m1Ado) and N7-methyl guanosine (m7Guo) in the presence of other nucleosides in urine. m1Ado as well as m7Guo were characterized by use of chromatographic techniques including substance specific chemical reactions.

tRNA modifications help maintain tRNA structure and facilitate translation and stress response. Found in all three kingdoms of life, m1A tRNA modification occurs in the T loop of many tRNAs, stabilizes tertiary tRNA structure, and impacts translation.

Methods
With the recent advanced direct RNA sequencing technique that proposed by the Oxford Nanopore Technologies, RNA modifications can be detected and profiled in a simple and straightforward manner. Majority nanopore-based modification studies were devoted to those popular types such as m6A and pseudouridine.
