Crystal structure of NAMPT with resistant mutations mapped.

open access: yes, 2014
The NAMPT protein structure is shown in ribbon diagram displaying an active dimer. The monomers of the NAMPT dimer are colored in brown and green, respectively.

Single-Cell RNA sequencing identifies NAMPT as a potential therapeutic target in autoimmune uveitis

open access: yesJournal of Advanced Research
Introduction: Autoimmune uveitis (AU) is an autoimmune disease of the eye that can lead to irreversible vision loss. Current therapies are limited by suboptimal efficacy and substantial side effects, highlighting the urgent need for the discovery of ...

TNFSF14/LIGHT responses in intestinal and oesophageal fibroblasts are differentially modulated by hydroxylase inhibitors

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend LIGHT is a mutual mediator of intestinal and oesophageal fibroblast inflammation with common as well as tissue‐specific effects. Hydroxylase inhibitors can selectively suppress inflammatory factors in both cell types, albeit targeting different signalling pathways.

Trophoblast Extracellular Vesicles Rewire Immunity: A Symptom‐Relief Avenue for Recessive Dystrophic Epidermolysis Bullosa

open access: yesJournal of Extracellular Vesicles, Volume 15, Issue 10, October 2026.
First‐trimester trophoblast EVs are enriched in anti‐inflammatory proteins, lipids, and immunoregulatory miRNAs. EVT‐EVs reprogram monocytes toward a pro‐resolving phenotype and restore immune homeostasis in RDEB, supporting their potential as offering a scalable off‐the‐shelf therapy for chronic inflammatory diseases.

Cancer cell metabolic plasticity allows resistance to NAMPT inhibition but invariably induces dependence on LDHA

open access: yesCancer & Metabolism, 2018
Background Inhibitors of nicotinamide phosphoribosyltransferase (NAMPT), the rate-limiting enzyme in NAD+ biosynthesis from nicotinamide, exhibit anticancer effects in preclinical models.

Brown Adipose Tissue: Molecular Mechanisms of Regulation, Physiological Functions, and Therapeutic Targets

open access: yesMedComm, Volume 7, Issue 10, October 2026.
This review systematically covers three core dimensions of brown adipose tissue (BAT): multilayered regulatory mechanisms governing brown adipocyte differentiation and phenotypic identity, physiological functions and signaling pathways in mediating thermogenesis and systemic metabolic crosstalk between BAT and multiple organs, and emerging clinical in ...

Targeting c-MYC through Interference with NAMPT and SIRT1 and Their Association to Oncogenic Drivers in Murine Serrated Intestinal Tumorigenesis

open access: yesNeoplasia: An International Journal for Oncology Research, 2019
We recently described a positive feedback loop connecting c-MYC, NAMPT, DBC1 and SIRT1 that contributes to unrestricted cancer cell proliferation. Here we determine the relevance of the loop for serrated route intestinal tumorigenesis using genetically ...

Securidaca inappendiculata Extract Mitigates Joint Damage in Collagen‐Induced Arthritis Rats Through Modulation of the SIRT1/HIF‐1α/Glycolysis Pathway

open access: yesPharmacology Research &Perspectives, Volume 14, Issue 5, October 2026.
ABSTRACT Securidaca inappendiculata (SI) Hassk. is a traditional Chinese herbal medicine commonly used in Southern China for the treatment of rheumatoid arthritis (RA). However, its exact mechanism of action remains unclear. In this study, we evaluated the therapeutic effects of the SI‐derived compound 1,7‐dihydroxy‐3,4‐dimethoxyxanthone (XAN) in a ...

Abstract LB-288: MPI-0486348: A novel inhibitor of nicotinamide phosphoribosyltransferase (Nampt) induces tumor regression in a preclinical model

open access: yes, 2010
Background: Nicotinamide phosphoribosyltransferase (Nampt) catalyzes the first step in the recycling of nicotinamide into NAD. Cancer cells develop dependence on Nampt due to increased energy requirements and the elevated activity of NAD consuming ...

Bi-directional crosstalk between NAD/NAMPT and IFN-γ/PD-L1axes in melanoma

open access: yes
Targeted therapy and immune checkpoint inhibitors (ICIs) have notably improved the treatment of BRAF- mutated metastatic melanoma (MM) patients; however, resistance mechanisms dramatically impact on the survival of patients.
