Ceramide‐mediated orchestration of oxidative stress response through filopodia‐derived small extracellular vesicles

open access: yesJournal of Extracellular Vesicles
Extracellular vesicles (EVs) are shed from the plasma membrane, but the regulation and function of these EVs remain unclear. We found that oxidative stress induced by H2O2 in Hela cells stimulated filopodia formation and the secretion of EVs.

ATRA transcriptionally induces nSMase2 through CBP/p300-mediated histone acetylation[S]

open access: yesJournal of Lipid Research, 2016
Neutral sphingomyelinase-2 (nSMase2) is a key ceramide-producing enzyme in cellular stress responses. While many posttranslational regulators of nSMase2 are known, emerging evidence suggests a more protracted regulation of nSMase2 at the transcriptional ...

Primary cilia in stem cells and neural progenitors are regulated by neutral sphingomyelinase 2 and ceramide

open access: yesMolecular Biology of the Cell, 2014
We show here that human embryonic stem (ES) and induced pluripotent stem cell–derived neuroprogenitors (NPs) develop primary cilia. Ciliogenesis depends on the sphingolipid ceramide and its interaction with atypical PKC (aPKC), both of which distribute to the primary cilium and the apicolateral cell membrane in NP rosettes.

Characterization of Neutral Sphingomyelinase Isoforms in Skeletal Muscle Mitochondria [PDF]

open access: yes, 2020
Skeletal muscle is composed of fiber types that differ in mitochondrial content, antioxidant capacity, and susceptibility to apoptosis. Ceramides have been linked to apoptosis by increasing mitochondrial permeability, a common hallmark of apoptotic ...

Adipocytes signal to recruit specific mRNAs from surrounding cells to restore expression deficits

open access: yesNature Communications
Extracellular vesicles (EVs) are nano-sized, membrane-delimited, particles released by cells that carry signaling macromolecules. A major pathway of EV production is potentiated by neutral sphingomyelinase 2 (SMPD3/nSMAse2), an enzyme that generates ...

Neutral sphingomyelinase 2 (nSMase2) is the primary neutral sphingomyelinase isoform activated by tumour necrosis factor-α in MCF-7 cells

open access: yes, 2011
Activation of N-SMase (neutral sphingomyelinase) is an established part of the response of cytokines such as TNF (tumour necrosis factor)-α. However, it remains unclear which of the currently cloned N-SMase isoforms (nSMase1, nSMase2 and nSMase3) are ...
