Methods to Activate the NLRP1 Inflammasome

2023
In addition to being the first NLR protein proposed to form inflammasome, NLRP1s have attracted much attention in their activation mechanism by post-translational auto-proteolysis to generate C-terminal CARD containing fragment to form inflammasome. Among NLRP1, mouse NLRP1B but not human NLRP1 is well studied for its activation by lethal toxin.

KSHV-encoded ORF45 activates human NLRP1 inflammasome

Nature Immunology, 2022
At steady state, the NOD-like receptor (NLR)-containing pyrin domain (PYD) (NLRP)1 inflammasome is maintained in an auto-inhibitory complex by dipeptidyl peptidases 8 and 9 (DPP8 and DPP9) and is activated by pathogen-encoded proteases after infection.

CARD8 negatively regulates NLRP1 inflammasome activation level by interaction with NLRP1

2022
AbstractNLRP1 inflammasome functions as canonical cytosolic sensor in response to intracellular infections and is implicated in auto-inflammatory diseases. But the regulation and signal transduction mechanisms of NLRP1 are incompletely understood. Here, we show that the T60 variant of CARD8, but not the canonical T48 isoform, negatively regulates NLRP1

Structural basis for thioredoxin-mediated suppression of NLRP1 inflammasome

Nature, 2023
Inflammasome sensors detect pathogen- and danger-associated molecular patterns and promote inflammation and pyroptosis1. NLRP1 was the first inflammasome sensor to be described, and its hyperactivation is linked to autoinflammatory disease and cancer2-6.

NLRP1‐associated autoinflammatory disease with epithelial dyskeratosis

Pediatric Dermatology, 2023
AbstractSeveral gain‐of‐function variants in NLRP1 cause a distinctive autoinflammatory disease reported under different names featuring mainly skin and mucosal involvement and variable systemic signs. Here, we report a new case of NLRP1‐associated autoinflammatory disease in a 6‐year‐old Peruvian girl, who presented with confluent hyperkeratotic ...

NEK7 activates the NLRP1 Inflammasome

2022
AbstractInflammasomes including those assembled by NLRP1 and NLRP3 regulate the innate immune system by inducing interleukin (IL)-1β and IL-18 maturation. Inflammasomes are functionally regulated by post-translational modifications such as phosphorylation.

Nlrp1 inflammasome is downregulated in trauma patients

Journal of Molecular Medicine, 2015
After a major trauma, IL-1β-producing capacity of monocytes is reduced. Generation of IL-1β is important for appropriate immune response after trauma and requires not only synthesis and transcription of inflammasome components but also their activation.

Reconstituting the NLRP1 Inflammasome In Vitro

2013
Nucleotide-binding domain leucine-rich-repeat containing receptors; NOD-Like Receptors (NLRs) were originally described as microbial sensors involved in host defense against pathogens that comprise an important component of the innate immune system. Recently, their cellular functions have expanded beyond classical pathogen detection, to danger sensors ...

Genetic variations ofNLRP1: susceptibility in psoriasis

British Journal of Dermatology, 2014
NACHT, LRR and PYD domain-containing protein (NLRP)1 is part of the inflammasome multiprotein complex involved in the production of interleukin (IL)-1β and IL-18, two cytokines strongly implicated in psoriasis pathogenesis. Genetic variations in NLRP1 are associated with a predisposition for chronic inflammatory conditions. The aim of the study was to ...

NALP1/NLRP1 Genetic Variants are Associated With Alzheimer Disease

Alzheimer Disease & Associated Disorders, 2012
Alzheimer disease (AD) is a complex neurodegenerative disease. Genetic and molecular studies have confirmed that in the human brain, amyloid-β fibrils can induce, through the activation of NALP1 inflammosome, inflammatory and apoptotic responses involved in the pathogenesis of AD. Considering that AD pathogenesis is multifactorial, we hypothesized that
