Orexin, Orexin Receptor Antagonists and Central Cardiovascular Control [PDF]
Orexin makes an important contribution to the regulation of cardiovascular function. When injected centrally under anesthesia, orexin increases blood pressure, heart rate and sympathetic nerve activity.

Evolution of Orexin Neuropeptide System: Structure and Function
Orexins are hypothalamic neuropeptides that were initially identified in the rat brain as endogenous ligands for an (previously) orphan G-protein-coupled receptor (GPCR).

Intra-periaqueductal Grey Matter Injection of Orexin A Attenuates Nitroglycerin-induced Deficits in Learning and Memory in Male Rats [PDF]
This study explored the potential contribution of Orx1R within vlPAG to the learning and memory of animals with chronic migraine-like pain. Migraine was induced by repeated i.p. administration of nitroglycerin (5 mg/kg).

Orexin B Reduces Cerebral Aneurysms Through Inhibition of SP-1. [PDF]
Orexin B/OX2R signaling attenuates cerebral aneurysm formation by suppressing SP‐1‐mediated inflammation, macrophage infiltration, and endothelial adhesion, with OX2R knockdown abolishing these protective effects. The study highlights Orexin B as a therapeutic candidate targeting vascular inflammation and remodeling in aneurysms.

Involvement of the Orexinergic System in Feeding
To know the processes involved in feeding, the dysregulation of hypothalamic neuropeptides promoting anorexigenic/orexigenic mechanisms must be investigated. Many neuropeptides are involved in this behavior and in overweight/obesity.

The neuropeptides orexins are important in regulating the neurobiological systems that respond to stressful stimuli. Furthermore, orexins are known to play a role many of the phenotypes associated with stress-related mental illness such as changes in cognition, sleep-wake states, and appetite.

Structural insights into the binding mode of the hypnotic drug vornorexant to orexin receptors. [PDF]
The crystal structure of the human orexin type 2 receptor–vornorexant complex (3.29 Å resolution) and orexin type 1 receptor docking revealed a conserved U‐shaped binding conformation, explaining the high affinity and balanced dual antagonism of this dual orexin receptor antagonist. The structural data validate the rational design that incorporated a 1,

Orexin Directly Excites Orexin Neurons through Orexin 2 Receptor [PDF]
Orexin neurons (hypocretin neurons) have a critical role in the regulation of sleep/wakefulness, especially in the maintenance of arousal. Here, we revealed that orexin neurons are directly and indirectly activated by orexin via the orexin 2 receptor (OX2R).

Research progress on the mechanism of orexin in pain regulation in different brain regions
Orexin is a neuropeptide that is primarily synthesized and secreted by the lateral hypothalamus (LH) and includes two substances derived from the same precursor (orexin A [OXA] and orexin B [OXB]).
