The E3 Ligase RNF8 Promotes Ubiquitination and Degradation of ChREBPα During Liver Stress Response

During MASH development, increasing hepatic stress signals, particularly a combination of lipotoxicity (palmitate) and inflammation (TNFα), trigger JNK, the key stress‐activated protein kinase. Activated JNK recruits RNF8, a stress‐induced E3 ligase, to promote ubiquitination and proteasomal degradation of the ChREBPα protein.

Saikosaponin A Inhibits Migration/Invasion and Triggers Cell‐Cycle Arrest and Autophagy‐Induced Cell Death in HCT116 Cells and In Vivo

Saikosaponin A from Radix Bupleuri inhibited colorectal cancer HCT116 cells with low toxicity. SSA induced S/G2‐M cell cycle arrest and triggered autophagy via mTOR inhibition. SSA suppressed metastasis by blocking PI3K/AKT signaling pathway. SSA significantly reduced tumor growth without apparent toxicity.
