Regulation of bile acid receptor activity

open access: yesLiver Research, 2018
Many receptors can be activated by bile acids (BAs) and their derivatives. These include nuclear receptors farnesoid X receptor (FXR), pregnane X receptor (PXR), and vitamin D receptor (VDR), as well as membrane receptors Takeda G protein receptor 5 ...

Review Article: Targeting Peroxisome Proliferator‐Activated Receptors in Primary Biliary Cholangitis

open access: yesAlimentary Pharmacology &Therapeutics, EarlyView.
Peroxisome proliferator‐activated receptor (PPAR) agonists have emerged as important second‐line treatments in primary biliary cholangitis, with profiles influenced by different selectivity for α‐, δ‐ and γ‐isoforms. PPAR‐α and PPAR‐δ agonism improve cholestasis, with inflammation reduced via all isoforms. PPAR‐δ agonism also reduces pruritus.

Novel plant inducers of PXR-dependent cytochrome P450 3A4 expression in HepG2 cells

open access: yesSaudi Pharmaceutical Journal, 2018
The cytochrome P450 3A4 (CYP3A4) is the most abundant CYP450 enzyme involved in the metabolism of endogenous products and xenobiotics, including prescription drugs and herbals.

Frequency of genetic polymorphisms of PXR gene in the Brazilian population [PDF]

open access: yes, 2011
INTRODUCTION: PXR polymorphisms have been implicated in modulating CYP3A4 and PXR expression, potentially accounting for interindividual differences in drug metabolism.

Promoter DNA methylation of farnesoid X receptor and pregnane X receptor modulates the intrahepatic cholestasis of pregnancy phenotype.

open access: yesPLoS ONE, 2014
The intrahepatic cholestasis of pregnancy (ICP) is a multifactorial liver disorder which pathogenesis involves the interplay among abnormal bile acid (BA) levels, sex hormones, environmental factors, and genetic susceptibility.

Regulation of xenobiotic and bile acid metabolism by the nuclear pregnane X receptor

open access: yesJournal of Lipid Research, 2002
The nuclear pregnane X receptor (PXR; NR1I2) is an integral component of the body's defense mechanism against chemical insult (chemoprotection). PXR is activated by a diverse array of lipophilic chemicals, including xenobiotics and endogenous substances,

A review of xenobiotic membrane transporter expression within the human placenta: Lessons gained from primary tissue and in vitro methodologies

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend A summary of the current knowledge around membrane transporter expression in the most widely used in vitro models of human trophoblasts, including primary placental tissue, cancer cell lines and trophoblast stem cells, as well as the techniques to attempt to mimic the human placenta in vivo.

The pregnane xenobiotic receptor, a prominent liver factor, has actions in the midbrain for neurosteroid synthesis and behavioral/neural plasticity of female rats [PDF]

open access: yes, 2014
A novel factor of interest for growth/plasticity in the brain is pregnane xenobiotic receptor (PXR). PXR is a liver factor known for its role in xenobiotic clearance and cholesterol metabolism.
