This study demonstrates a cuproptosis mechanism involving nicotinamide adenine dinucleotide (NADH)‐reductive stress in neural cells. Copper activates dihydrolipoamide dehydrogenase under mitochondrial pH, accumulating NADH. Copper also induces mitochondrial permeability transition pore opening, facilitating NADH translocation to the cytosol and ...

CoA-dependent activation of mitochondrial acyl carrier protein links four neurodegenerative diseases [PDF]
PKAN, CoPAN, MePAN, and PDH-E2 deficiency share key phenotypic features but harbor defects in distinct metabolic processes. Selective damage to the globus pallidus occurs in these genetic neurodegenerative diseases, which arise from defects in CoA ...

Diversity of FAAL enzymes and prediction of their substrate specificity using FAALPred
Abstract FAALs (fatty acyl‐AMP ligases) recruit and incorporate fatty acids during the biosynthesis of secondary metabolites. Their diversity, distribution, and substrate specificity remain poorly understood, which limits functional predictions from sequence data.

Ohr (Organic Hydroperoxide Resistance Protein) Possesses a Previously Undescribed Activity, Lipoyl-dependent Peroxidase [PDF]
The Ohr (organic hydroperoxide resistance) family of 15-kDa Cys-based, thiol-dependent peroxidases is central to the bacterial response to stress induced by organic hydroperoxides but not by hydrogen peroxide. Ohr has a unique three-dimensional structure and requires dithiols, but not monothiols, to support its activity.

Boosting Cuproptosis in Breast Cancer Therapy via Photodynamic Treatment With a New Liposome
ABSTRACT Breast cancer (BC) is one of the most common cancers in women around the world, and utilizing a combined approach is a crucial strategy. Induction of cuproptosis in tumor cells is a novel antitumor approach, though its standalone efficacy remains unclear.

Progress of Programmed Cell Death in Steroid‐Induced Osteonecrosis of the Femoral Head
Steroid‐induced osteonecrosis of the femoral head is a severe osteoarticular condition resulting from glucocorticoid overuse, characterized by femoral head bone structure collapse and cell death, now predominant among nontraumatic femoral head necroses.

Lipidomic Analysis of Plastidial Octanoyltransferase Mutants of Arabidopsis thaliana [PDF]
International audiencePlant de novo fatty acid synthesis takes place in the plastid using acetyl-coenzyme A (acetyl-CoA) as the main precursor. This first intermediate is produced from pyruvate through the action of the plastidial pyruvate dehydrogenase ...

Mitochondrial Fatty Acids and Neurodegenerative Disorders [PDF]
Fatty acids in mitochondria, in sensu stricto, arise either as β-oxidation substrates imported via the carnitine shuttle or through de novo synthesis by the mitochondrial fatty acid synthesis (mtFAS) pathway. Defects in mtFAS or processes involved in the

Metabolic reorganization and chronic inflammation are dynamic and diverse events in the tumor microenvironment (TME) that cause tumor propagation, spreading, and resistance to treatment. However, copper‐induced cell death (cuproptosis), which is a newly discovered form of regulated cell death (RCD) induced by high intracellular copper ions and is a ...
