Control of protein synthesis through mRNA pseudouridylation by dyskerin

open access: yesScience Advances, 2023
Posttranscriptional modifications of mRNA have emerged as regulators of gene expression. Although pseudouridylation is the most abundant, its biological role remains poorly understood. Here, we demonstrate that the pseudouridine synthase dyskerin associates with RNA polymerase II, binds to thousands of mRNAs, and is responsible for their ...

Unraveling the mechanisms of pseudouridylation in rice plant immunity: The power of next-generation sequencing

open access: yesCurrent Plant Biology
Pseudouridylation is a prevalent RNA modification that plays a pivotal role in regulating gene expression and modulating plant responses to biotic stresses, such as pathogenic attacks.

Inhibition of human dyskerin as a new approach to target ribosome biogenesis. [PDF]

open access: yesPLoS ONE, 2014
The product of the DKC1 gene, dyskerin, is required for both ribosome biogenesis and telomerase complex stabilization. Targeting these cellular processes has been explored for the development of drugs to selectively or preferentially kill cancer cells ...

Targeting PUS7 suppresses tRNA pseudouridylation and glioblastoma tumorigenesis

open access: yesNature Cancer, 2021
Pseudouridine is the most frequent epitranscriptomic modification. However, its cellular functions remain largely unknown. Here, we show that pseudouridine synthase 7 (PUS7) is highly expressed in glioblastoma versus normal brain tissues, and high PUS7 expression levels are associated with worse survival in patients with glioblastoma.

Overview on Epstein–Barr Virus–Host Interactions With a Focus on Host Cell Transcriptome Alterations

open access: yesCell Biochemistry and Function, Volume 44, Issue 10, October 2026.
ABSTRACT The Epstein–Barr virus (EBV) belongs to the gamma subgroup of human herpesviruses. EBV is particularly lymphotropic regarding its latency, meaning that it preferentially targets lymphocytes as its immunological niche. During the establishment of the complex virus–host relationships, the virus influences the human host cell transcriptome ...

How Epitranscriptomic Machinery Senses Environmental Cues

open access: yesAdvanced Biology, Volume 10, Issue 9, September 2026.
Exogenous exposures, microenvironmental signals, and intracellular metabolic or redox states can alter RNA‐modification machinery through catalytic chemistry, signaling‐dependent PTMs, compartmental redistribution, or changes in abundance and composition.

SNORA56-mediated pseudouridylation of 28 S rRNA inhibits ferroptosis and promotes colorectal cancer proliferation by enhancing GCLC translation

open access: yesJournal of Experimental & Clinical Cancer Research, 2023
Background Colorectal cancer (CRC) is one of the most common malignancies and is characterized by reprogrammed metabolism. Ferroptosis, a programmed cell death dependent on iron, has emerged as a promising strategy for CRC treatment.

Epitranscriptomic Regulation of Bone Metabolism: Emerging Roles of N6‐Methyladenosine (m6A) Modification in Skeletal Homeostasis

open access: yesOral Science International, Volume 23, Issue 3, September 2026.
ABSTRACT N6‐methyladenosine (m6A) is the most abundant internal chemical modification of eukaryotic RNA and a central mechanism of epitranscriptomic gene regulation. Although m6A was first characterized in messenger RNA (mRNA), it is also deposited on ribosomal RNA (rRNA), circular RNA, and other RNA species, where it regulates RNA stability, decay ...

RNA‐Protein Assemblies: A Review of Biophysical Principles and Coarse‐Grained Modeling Approaches

open access: yesWIREs RNA, Volume 17, Issue 5, September/October 2026.
RNA‐protein assemblies in the nucleolus, nuclear speckles, and viral ribonucleoprotein complexes are governed by multivalent IDR‐RNA interactions that drive liquid–liquid phase separation. This review examines the molecular grammar underlying these systems and critically evaluates coarse‐grained computational frameworks for modeling their sequence ...

Insight into the mechanisms and functions of spliceosomal snRNA pseudouridylation

open access: yesWorld Journal of Biological Chemistry, 2014
Pseudouridines (Ψs) are the most abundant and highly conserved modified nucleotides found in various stable RNAs of all organisms. Most Ψs are clustered in regions that are functionally important for pre-mRNA splicing. Ψ has an extra hydrogen bond donor that endows RNA molecules with distinct properties that contribute significantly to RNA-mediated ...
