Olfactory tuft cells communicate with other mucosal cells in maintaining olfactory epithelial immunity and sensory function. Disruption of tuft cell signalling pathways causes a skewed immune response, leading to heightened inflammation and increased activation of the quiescent stem cells, which are further exacerbated by viral infection.

SOX4-STAT6-MTHFD2 axis drives hepatocellular carcinoma progression and treatment resistance
Hepatocellular carcinoma (HCC) is a major global health burden. Despite recent advances in immunotherapy, tyrosine kinase inhibitors (TKIs) treatment or combined therapies, therapeutic resistance and disease progression remain significant challenges ...

NAB2-STAT6 drives an EGR1-dependent neuroendocrine program in solitary fibrous tumors
The pathogenesis of many rare tumor types is poorly understood, preventing the design of effective treatments. Solitary fibrous tumors (SFTs) are neoplasms of mesenchymal origin that affect 1/1,000,000 individuals every year and are clinically ...

From hepatic to hematopoietic: LRH‐1's expanding cellular repertoire to the immune system
The nuclear receptor LRH‐1 is a well‐characterized regulator of endodermal tissue. Yet, increasing evidence indicates that LRH‐1, although expressed at low levels, is also a critical regulator of the hematopoietic system. LRH‐1 regulates the immune system by contributing to immune cell‐specific functions.

Statistical reconstruction of transcription factor activity using Michaelis-Menten kinetics [PDF]
Summary. The basic building block of a gene regulatory network consists of a gene encoding a transcription factor (TF) and the gene(s) it regulates. Considerable efforts have been directed recently at devising experiments and algorithms to determine TFs

Chemotherapy is the leading treatment for acute lymphoblastic leukemia (ALL). However, many ALL patients eventually develop relapses, the treatment of which remains a major challenge due to their chemoresistance phenotype.

During homeostasis, CCL22 is secreted by dendritic cells (DC), which mediate contacts between regulatory T cells (Treg) and DC to maintain immune balance. Activation of innate immunity via microbes or pattern recognition receptor (PRR) agonists induces the secretion of several soluble factors (e.g., type I and II interferons, IL‐10), which mediate ...

Clostridioides difficile toxins TcdA and TcdB enter intestinal epithelial cells (IECs) via specific receptors, disrupt tight‐junction integrity and activate PI3K/Akt/mTOR signalling, promoting inflammasome assembly and IL‐1β release. Toxin‐induced IEC apoptosis further weakens the epithelial barrier.

Emerging Roles of PSTPIP2 in Autoimmunity: From Mechanism to Therapeutic Implications
PSTPIP2 acts as a key brake on myeloid inflammation by suppressing IL‐1β‐driven signalling, macrophage dysregulation and inflammation‐associated bone remodelling. Its emerging roles in CRMO/CMO, rheumatoid arthritis, SAPHO syndrome and bullous pemphigoid support its potential as a biomarker and therapeutic target.

Innovations in Obesity Treatment: Beyond Adipose Tissue Dysfunction
Obesity drives chronic inflammation, insulin resistance, type 2 diabetes, and cancer development through adipocyte dysfunction. Addressing this multisystemic disorder requires integrated strategies beyond diet and exercise, such as thermogenesis activation via menthol or capsinoids and appetite control through GLP‐1/GIP agonists and neuromodulation to ...
