In nonribosomal peptide synthetases, the peptidyl‐carrier‐protein (PCP) of each module tethers the amino acid to be incorporated into the peptide product. Its interaction with multiple catalytic domains is barely studied. We reveal a surprising competition of the adenylation (A) and epimerization (E) domains for the PCP in an A‐PCP‐E module.
