T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome

A model of tertiary lymphatic structure-related prognosis for penile squamous cell carcinoma
Background We investigated the feasibility of the tertiary lymphoid structure (TLS) as a prognostic marker for penile squamous cell carcinoma(SCC). Methods We retrospectively collected data from 83 patients with penile squamous cell carcinoma.

Autologous tumor‐infiltrating lymphocyte (TIL) therapy shows promising efficacy in acral melanoma, yet determinants of durable response remain unclear. By integrating single‐cell transcriptomics and TCR sequencing, this study reveals that TIL products enriched for T follicular helper and intermediate exhausted T cells establish persistent clonal ...

Abstract Colorectal cancer (CRC) ranks as the third most common and second deadliest cancer worldwide. The immune system, particularly tertiary lymphoid structures (TLS), significantly influences CRC progression and prognosis. TLS maturation, especially in the presence of germinal centers, correlates with improved patient outcomes; however ...

ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.

Radiation drives tertiary lymphoid structures to reshape TME for synergized antitumour immunity
Radiotherapy (RT) plays a key role in the tumour microenvironment (TME), impacting the immune response via cellular and humoral immunity. RT can induce local immunity to modify the TME.

SSR4, a TRAP component induced in B cells, governs BAFFR N‐glycosylation via DDOST to sustain NF‐κB signaling, B‐cell differentiation, and TLS maturation. Its loss impairs anti‐tumor immunity, while overexpression improves antibody glycosylation and ADCC, revealing a critical regulator for cancer immunotherapy.
