The E3 ubiquitin ligase STUB1 regulates autophagy and mitochondrial biogenesis by modulating TFEB activity

open access: yesMolecular & Cellular Oncology, 2017
TFEB is a master regulator for transcription of genes involved in autophagy, lysosome and mitochondrial biogenesis. Activity of TFEB is inhibited upon its phosphorylation.

TFEB‐dependent lysosome biogenesis is required for senescence

open access: yesThe EMBO Journal, 2023
Abstract The accumulation of senescent cells is recognised as a driver of tissue and organismal ageing. One of the gold‐standard hallmarks of a senescent cell is an increase in lysosomal content, as measured by senescence‐associated β‐galactosidase (Senβ‐Gal) activity.

Transcriptional factor EB regulates macrophage polarization in the tumor microenvironment

open access: yesOncoImmunology, 2017
Tumor microenvironment (TME) contains a variety of infiltrating immune cells. Among them, tumor-associated macrophages (TAMs) and their alternative activation contribute greatly to the progression of tumors.

TFEB-driven endocytosis coordinates MTORC1 signaling and autophagy

open access: yes, 2018
The mechanistic target of rapamycin kinase complex 1 (MTORC1) is a central cellular kinase that integrates major signaling pathways, allowing for regulation of anabolic and catabolic processes including macroautophagy/autophagy and lysosomal biogenesis ...

TFEB-induced lysosome biogenesis inhibits C. burnetii growth.

open access: yes, 2019
(A) Representative images of immunofluorescence staining of WT C. burnetii-infected parental and TFEB-GFP HeLa cells. Fixed cells were stained for C. burnetii and CD63, a CCV marker. Arrows point to individual CCVs.

TFEB-dependent induction of thermogenesis by the hepatocyte SLC2A inhibitor trehalose

open access: yes, 2018
The macroautophagy/autophagy-inducing disaccharide, trehalose, has been proposed to be a promising therapeutic agent against neurodegenerative and cardiometabolic diseases.

Autophagy and mitophagy in pancreatic β‐cell homeostasis and their involvement in diabetes pathophysiology

open access: yesFEBS Letters, EarlyView.
This review focuses on the role of autophagy and mitophagy in maintaining pancreatic β‐cell function and homeostasis. We discuss how genetic defects affecting these pathways contribute to the development of type 1, type 2, monogenic, and gestational diabetes. We further explore their potential as therapeutic targets. Created in BioRender.

Cell surface CD11c as a neutrophil aging marker molecule

open access: yesFEBS Open Bio, EarlyView.
Cell surface CD11chi neutrophils were more aged and had better phagocytic function than CD11c−/lo neutrophils. Transcriptomic analysis of CD11chi neutrophils and CD11c−/lo neutrophils in pediatric population showed that the most difference was seen in infants.

STUB1 regulates TFEB-induced autophagy-lysosome pathway

open access: yes, 2017
TFEB is a master regulator for transcription of genes involved in autophagy and lysosome biogenesis. Activity of TFEB is inhibited upon its serine phosphorylation by mTOR The overall mechanisms by which TFEB activity in the cell is regulated are not well

TRPML1 agonist ML‐SA5 attenuates pulmonary fibroblast activation by suppressing mTOR and restoring autophagic flux

open access: yesFEBS Open Bio, EarlyView.
TGF‐β1 stimulation downregulates lysosomal channel TRPML1 in pulmonary fibroblasts. The TRPML1 agonist ML‐SA5 suppressed fibroblast‐to‐myofibroblast activation and collagen production. Mechanistically, ML‐SA5 inhibited mTOR phosphorylation, restored autophagic flux, and its effects were enhanced by rapamycin (mTOR inhibitor) and reversed by MHY1485 ...
