Targeting PLD3 Reverses the Immunosuppressive Niche by Reprogramming Tumor‐Associated Macrophages and Potentiates Antitumor Immunity

open access: yesAdvanced Science, EarlyView.
PLD3 activates the lysosomal‐AKT‐NF‐κB axis to drive cellular senescence in macrophages, establishing an immunosuppressive TME by limiting the infiltration of cytotoxic T, NK, and NKT cells, which confers resistance to anti‐PD‐1 therapy. Abrine inhibits PLD3 expression, restoring antitumor immunity and synergizing with anti‐PD‐1 treatment.

Differential regulation of Treg stability in human naïve and effector Treg subsets by TGFβ-signaling via ARKADIA-SKI axis [PDF]

open access: yes
IntroductionThe human FOXP3+ regulatory T (Treg) cells, a subset of CD4+ T cells with immunosuppressive function, are essential for the maintenance of immune homeostasis and tolerance.

Reprogramming the Immune Landscape of Inflammatory Breast Cancer

open access: yesAdvanced Science, EarlyView.
Inflammatory breast cancer is the most lethal subtype of breast cancer and is characterized by an immunosuppressive tumor microenvironment (TME) driven by a complex network of immune cells and inflammatory cytokines, contributing to its aggressiveness and treatment challenges. Immune checkpoint inhibitors, either alone or in combination, show potential

Interaction between Treg apoptosis pathways, Treg function and HLA risk evolves during type 1 diabetes pathogenesis.

open access: yes, 2012
We have previously reported increased apoptosis of regulatory T cells (Tregs) in recent-onset Type 1 Diabetes subjects (RO T1D) in the honeymoon phase and in multiple autoantibody-positive (Ab+) subjects, some of which are developing T1D.

PGK1 Suppresses CD8+ T Cell‐Mediated Antitumor Immunity Through CCL2/CCR2/Tumor‐Associated Macrophages Axis in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
PGK1 in tumor cells upregulates CCL2 expression through activation of the AKT/GSK‐3β/β‐catenin signaling axis, thereby promoting the recruitment and M2 polarization of TAMs and ultimately impairing the infiltration and activation of CD8+ T cells within the HCC tumor microenvironment. ABSTRACT Patients with advanced hepatocellular carcinoma (HCC) have a

Improved migration of tumor ascites lymphocytes to ovarian cancer microenvironment by CXCR2 transduction

open access: yesOncoImmunology, 2018
Chemokines are essential mediators of cellular trafficking, interactions and tumor development. Though adoptive cell therapy (ACT) has been a tremendous success in the treatment of metastatic melanoma (MM), a major obstacle for successful ACT, is limited

Expression of IFN-γ, TNF-α, IL17 in Treg of PBLs or in expanded Treg.

open access: yes, 2013
Cells from PBMCs and expanded Treg on Day 14 after anti-CD3/CD28 activation were stained with indicated antibodies following stimulation with PMA/ionomycin for 5 h and analyzed by FACS.

Artificial Intelligence‐Based Body Composition Analysis Reveals Sex‐Specific Prognostic Markers and Their Clinical Value in Gastric Cancer: A Multicenter Study

open access: yesAdvanced Science, EarlyView.
This study applied AI to quantify multidimensional body composition from CT images in gastric cancer and healthy controls. Distinct sex‐specific patterns and disease‐related alterations were identified and were associated with survival. Higher muscle and fat measures were linked to improved outcomes.

Heat Shock Protein 90: From Molecular Chaperone Function to Therapeutic Targeting in Malignancies

open access: yesAdvanced Science, EarlyView.
In this review, an integrated conceptual framework linking HSP90's molecular chaperone functions to its pathological roles in cancer is proposed. HSP90 serves as a central node that integrates oncogenic signaling, buffers proteotoxic stress, maintains cancer stem cell plasticity, and shapes tumor‐immune interactions, all of which converge to drive ...
