Crystallization of α-Galactosidase from Trichoderma reesei

Journal of Molecular Biology, 1993
An extracellular alpha-galactosidase was isolated from fungus Trichoderma reesei. The purified enzyme had a molecular weight of 54,000 Da and an isoelectric point of 5.25. Crystals of alpha-galactosidase were obtained from a polyethylene glycol 4000 solution by the hanging-drop method. Seeding was used for enlargement of the crystal size.

Sphaeroplast formation and regeneration in Trichoderma reesei

European Journal of Applied Microbiology and Biotechnology, 1981
Sphaeroplasts from several genetically marked strains of Trichoderma reesei were readily prepared through enzymatic hydrolysis of mycelial suspensions utilizing Driselase, a commercially available lytic enzyme preparation. Sphaeroplasts were released from the apical tips of hyphae after 90 min exposure to the enzyme and, with longer treatments, from ...

Cellulases of Trichoderma reesei

1988
Publisher Summary Trichoderma reesei QM6a and a mutant from this strain produce very effective cellulases. The protein exocellobiohydrolas (CBH 1) is the most abundant of the enzymes from Trichoderma reesei . Trichoderma reesei also produces cellobiase activity.

Natural products isolation from Trichoderma reesei

2022
The focus of the present work is to develop Trichoderma reesei as a heterologous host for the production of natural products and to explore the biosynthetic potential of this fungus by expressing one of its cryptic PKS-NRPS gene clusters. Developing T.

In Vivo Footprinting Analysis in Trichoderma reesei

2020
The in vivo footprinting method identifies protein-targeted DNA regions under different conditions such as carbon sources. Dimethyl sulfate (DMS) generates methylated purine bases at DNA sites which are not bound by proteins or transcription factors.

Proteomics studies in Trichoderma reesei

2002
Proteome analysis is the simultaneous examination of a large subset of the protein species in a given cell. Traditionally proteomics is performed by two-dimensional gel electrophoresis combined with protein identification by mass spectrometry. We have set up proteomics methodology for the analysis of both extracellular and intracellular proteins of ...

Promoters and Synthetic Promoters in Trichoderma reesei

Trichoderma reesei holds immense promise for large-scale protein production, rendering it an excellent subject for deeper exploration using genetic engineering methods to achieve a comprehensive grasp of its cellular physiology. Understanding the genetic factors governing its intrinsic regulatory network is crucial, as lacking this knowledge could ...

Proteome analysis in Trichoderma reesei

2004
We have set up methods for intracellular and extracellular proteome analysis in Trichoderma. These include analysis of steady state total protein pattern using 2D gel electrophoresis as well as detection of actively synthesised proteins using metabolic labelling.
