In Vitro Reconstruction of Axonal Heat Sensing with a Photothermal Nerve‐on‐a‐Chip

A photothermal nerve‐on‐a‐chip platform is presented that enables precise, localized heating of sensory axons with simultaneous electrical recording. The system reveals millisecond‐scale dynamics of heat‐evoked neural activity and provides a new approach to study how temperature‐dependent ion channel behavior is translated into axonal firing.

TRPA1 Antagonists for Pain Relief [PDF]

Here, we review the literature assessing the role of transient receptor potential ankyrin 1 (TRPA1), a calcium-permeable non-selective cation channel, in various types of pain conditions. In the nervous system, TRPA1 is expressed in a subpopulation of nociceptive primary sensory neurons, astroglia, oligodendrocytes and Schwann cells.

Ligustilide: a novel TRPA1 modulator

TRPA1 is activated by electrophilic compounds such as mustard oil (MO). Here, we demonstrate a bimodal sensitivity of TRPA1 to ligustilide (Lig), an electrophilic volatile dihydrophthalide of dietary and medicinal relevance. Lig is a potent TRPA1 activator and is also capable to induce a modest block of MO activated currents.

Distinctly altered TRPC3 and TRPC6 expression patterns in human Alzheimer's disease cortex and hippocampus

Divergent remodeling of TRPC channels occurs in Alzheimer's disease, with TRPC3 increased in neurons and reactive astrocytes and TRPC6 reduced in pyramidal neurons. These alterations may contribute to calcium dysregulation, neuroinflammation, and synaptic dysfunction during AD progression.
