Small ubiquitin-like modifier protein SUMO enables plants to control growth independently of the phytohormone gibberellin [PDF]

open access: yes, 2014
Plants survive adverse conditions by modulating their growth in response to a changing environment. Gibberellins (GAs) play a key role in these adaptive responses by stimulating the degradation of growth-repressing DELLA proteins.

LKB1 Dysregulation in Duchenne Muscular Dystrophy Models: Disease Specificity and Epigenetic Control by HDAC Inhibitors

open access: yesAnnals of the New York Academy of Sciences, Volume 1562, Issue 1, August 2026.
Our study demonstrated that the LKB1–STRADα–MO25 complex was significantly downregulated in Duchenne muscular dystrophy (DMD) models, concurring with the disruption of mechano‐metabolic signaling. Treatment with the HDAC inhibitor, vorinostat, successfully restored LKB1 expression by suppressing specific repressor miRNAs.

A ubiquitin-specific protease that efficiently cleaves the ubiquitin-proline bond.

open access: yes, 2022
Ubiquitin is a small eukaryotic protein that is synthesized naturally as one of several fusion proteins, which are processed by ubiquitin-specific proteases to release free ubiquitin.

Covalent Inhibition of SHMT2 by Gambogic Acid Induces Ferroptosis Through Mitochondrial Collapse in Triple‐Negative Breast Cancer

open access: yesAdvanced Science, Volume 13, Issue 42, 28 July 2026.
In this study, chemoproteomics combined with genetic and functional analyses was integrated to identify SHMT2 as a covalent and functional target of gambogic acid (GA) in triple‐negative breast cancer (TNBC). Further validation demonstrated that GA selectively modifies the Cys241 site of SHMT2, triggering mitochondrial dysfunction, activating the Nrf2 ...

SUMO chain-induced dimerization activates RNF4 [PDF]

open access: yes, 2014
Dimeric RING E3 ligases interact with protein substrates and conformationally restrain the ubiquitin-E2-conjugating enzyme thioester complex such that it is primed for catalysis.

USPs and non-coding RNAs: Unraveling their interplay within tumor microenvironment for targeted cancer therapy

open access: yesPharmacological Research
Cancer represents a significant global health threat, with the World Health Organization reporting millions of new cases annually. Dysregulation in cellular homestasis leads to the aggressive and heterogeneous nature of cancers.

Discovery of a Potent Fluorescence Polarization Probe for Identifying USP1 Allosteric Inhibitors

open access: yesAdvanced Science, Volume 13, Issue 39, 13 July 2026.
This study presents the first ubiquitin‐specific protease 1 (USP1) allosteric fluoroprobe and fluorescence polarization assay, enabling the differentiation of allosteric and catalytic site inhibitors. Further, a novel class of tetrahydroisoquinoline‐based USP1 inhibitors is designed, with compound 14a (USP1 IC50 = 29.9 nM) showing strong selectivity ...

Non‐canonical function of transferrin receptor‐1 promotes breast cancer metastasis by activating HCK‒STAT3‒MMP9 signalling

open access: yesClinical and Translational Medicine, Volume 16, Issue 7, July 2026.
Schematic summary of TfR‐1‐induced HCK axis activation promoting breast cancer metastasis: TfR‐1 interacts with USP32 and stabilises HCK, directly mediating its phosphorylation and activation. Activated HCK subsequently induces STAT3 phosphorylation and nuclear accumulation, which drives transcriptional upregulation of MMP9, a key mediator of ...

Chemistry-based functional proteomics: mechanism-based activity-profiling tools for ubiquitin and ubiquitin-like specific proteases.

open access: yes, 2004
Determining the biological function of newly discovered gene products requires the development of novel functional approaches. To facilitate this task, recent developments in proteomics include small molecular probes that target proteolytic enzyme ...

Ubiquitin and ubiquitin-like specific proteases targeted by infectious pathogens: Emerging patterns and molecular principles. [PDF]

open access: yes, 2008
Attachment of ubiquitin (Ub) or ubiquitin-like (Ubl) modifiers is a reversible post-translational modification that regulates the fate and function of proteins.
