Transient Interdomain Interactions Modulate the Monomeric Structural Ensemble and Self‐Assembly of Huntingtin Exon 1

open access: yesAdvanced Science, EarlyView.
Polyglutamine (polyQ) tract expansion (≥ 36 amino acids) within the N‐terminal region of the Huntingtin protein (Httex1) causes Huntington's disease (HD), for which the underlying causes are not well‐understood. The authors performed computer simulations to understand the cause of HD at the molecular level.

Atrial Natriuretic Peptide Orchestrates a Coordinated Physiological Response to Fuel Non-shivering Thermogenesis

open access: yesCell Reports, 2020
Summary: Atrial natriuretic peptide (ANP) is a cardiac hormone controlling blood volume and pressure in mammals. It is still unclear whether ANP controls cold-induced thermogenesis in vivo.

FGF6 and FGF9 regulate UCP1 expression independent of brown adipogenesis

open access: yesNature Communications, 2020
Uncoupling protein-1 (UCP1) plays a central role in energy dissipation in brown adipose tissue. Here the authors show that FGF6 and FGF9 induce UCP1 expression in adipocytes and preadipocytes, via modulation of a transcriptional network that is ...

Telmisartan prevents weight gain and obesity through activation of peroxisome proliferator-activated receptor-delta-dependent pathways [PDF]

open access: yes, 2010
—Telmisartan shows antihypertensive and several pleiotropic effects that interact with metabolic pathways. In the present study we tested the hypothesis that telmisartan prevents adipogenesis in vitro and weight gain in vivo through activation of ...

Cardiac Slc25a49‐Mediated Energy Reprogramming Governs Doxorubicin‐Induced Cardiomyopathy through the G6P–AP‐1–Sln Axis

open access: yesAdvanced Science, EarlyView.
Doxorubicin‐induced cardiomyopathy involves mitochondrial energy metabolism dysfunction, exacerbated by cardiomyocyte‐specific Slc25a49 deficiency via oxidative phosphorylation (OXPHOS) suppression and glycolysis activation. The Slc25a49–glucose‐6‐phosphate (G6P)–activator protein‐1 (AP‐1) axis drives myocardial injury by upregulating Sln, disrupting ...

Endogenous FGF21-signaling controls paradoxical obesity resistance of UCP1-deficient mice

open access: yesNature Communications, 2020
Brown adipose thermogenesis increases energy expenditure and relies on uncoupling protein 1 (UCP1), however, UCP1 knock-out mice show resistance to diet-induced obesity at room temperature.

Amino Acid‐Sensing Neurons in the Anterior Piriform Cortex Control Brown Adipose Tissue Thermogenesis

open access: yesAdvanced Science, EarlyView.
This study identifies a class of amino acid‐sensing neurons, APCCRH neurons, which regulate BAT thermogenesis. Leucine deficiency alters intrinsic excitability through GCN2 signaling pathway, activating APCCRH neurons, which in turn regulate BAT thermogenesis via projections to the lateral hypothalamus.

Anti-oxidant and anti-inflammatory activity of ketogenic diet. New perspectives for neuroprotection in alzheimer’s disease [PDF]

open access: yes, 2018
The ketogenic diet, originally developed for the treatment of epilepsy in non-responder children, is spreading to be used in the treatment of many diseases, including Alzheimer’s disease.

Mechanical Stress Induced NOX2 Promotes Endothelial Dysfunction in Ventilator‐Induced Lung Injury: Potential Treatment with Quercetin

open access: yesAdvanced Science, EarlyView.
Abnormal mechanical stretch induced by mechanical ventilation (MV) disrupts endothelial junctions via the NOX2/ROS/CaMKII/ERK1/2 axis both in human aortic endothelial cells (HAECs) in vitro and in ventilator‐induced lung injury (VILI) in mice. Quercetin, an antioxidant predicted to bind to NOX2, can effectively prevent mechanical stretch‐induced ...

Uncoupling protein-1 as a target for the treatment of obesity/insulin resistance

open access: yesFrontiers in Physiology, 2015
Presence of brown adipose tissue (BAT), characterised by the expression of the thermogenic uncoupling protein 1 (UCP1), has recently been described in adult humans.
