Roles of Wapl in cell cycle progression and chromosome segregation

Sister chromatid cohesion on chromosome arms is essential for the segregation of homologous chromosomes during meiosis I while it is dispensable for sister chromatid separation during mitosis.

Maintenance and precise regulation of sister chromatid cohesion is essential for faithful chromosome segregation during mitosis and meiosis. Cohesin cofactors contribute to cohesin dynamics and interact with cohesin complexes during cell cycle.

ESCO1 and CTCF enable formation of long chromatin loops by protecting cohesinSTAG1 from WAPL.
Eukaryotic genomes are folded into loops. It is thought that these are formed by cohesin complexes via extrusion, either until loop expansion is arrested by CTCF or until cohesin is removed from DNA by WAPL.

Wapl antagonizes cohesin binding and promotes Polycomb-group silencing inDrosophila
Wapl protein regulates binding of the cohesin complex to chromosomes during interphase and helps remove cohesin from chromosomes at mitosis. We isolated a dominant mutation in wapl (waplAG) in a screen for mutations that counteract silencing mediated by an engrailed Polycomb-group response element.

Sister centromere fusion is a process unique to meiosis that promotes co-orientation of the sister kinetochores, ensuring they attach to microtubules from the same pole during metaphase I.

Investigation of the role of Pds5 and Wapl in sister chromatid separation in mammalian cells
The key element of sister chromatid cohesion (SCC) is the cohesin complex composed of Smc1, Smc3, Scc1 and Scc3 (SA1 and SA2 in vertebrates). However, additional proteins such as Pds5 and Wapl are also known to regulate SCC. Most of our knowledge of Pds5

CTCF, WAPL and PDS5 proteins control the formation of TADs and loops by cohesin
Mammalian genomes are organized into compartments, topologically-associating domains (TADs) and loops to facilitate gene regulation and other chromosomal functions. Compartments are formed by nucleosomal interactions, but how TADs and loops are generated

For self-renewal, embryonic stem cells (ESCs) require the expression of specific transcription factors accompanied by a particular chromosome organization to maintain a balance between pluripotency and the capacity for rapid differentiation. However, how
