CYP46A1 activation by low-dose efavirenz enhances brain cholesterol metabolism in subjects with early Alzheimer’s disease

open access: yesAlzheimer’s Research & Therapy, 2022
Background Efavirenz is an anti-HIV drug, and cytochrome P450 46A1 (CYP46A1) is a CNS-specific enzyme that metabolizes cholesterol to 24-hydroxycholesterol (24HC).

Changes in the gut microbiome during fasting‐induced molting and its effects on gut–liver function

open access: yesiMetaOmics, Volume 3, Issue 2, June 2026.
During fasting, harmful microbiota and metabolites rise, triggering intestinal injury and activation of the TGF‐β signaling pathway, which enhances intestinal stem cell proliferation. Concurrently, hepatic function declines, indicated by increased hepatic and serum bile acids and upregulation of the non‐classical bile acid synthesis pathway.

Diagnostic Power of 24S-Hydroxycholesterol in Cerebrospinal Fluid: Candidate Marker of Brain Health

open access: yesJournal of Alzheimer’s Disease, 2013
We evaluated the diagnostic potential of 24S-hydroxycholesterol (24OHC) in cerebrospinal fluid. At a memory clinic, we investigated subjects with subjective cognitive impairment (SCI, n = 33), mild cognitive impairment (MCI) patients (n = 27), MCI patients with later progression into Alzheimer dementia at follow up (n = 10), and ...

24S-hydroxycholesterol effects on lipid metabolism genes are modeled in traumatic brain injury [PDF]

open access: yesBrain Research, 2010
Membrane damage during traumatic brain injury (TBI) alters the brain homeostasis of cholesterol and other lipids. Cholesterol 24S-hydroxylase (Cyp46) is a cholesterol metabolic enzyme that is increased after TBI. Here, we systematically examined the effects of the enzymatic product of Cyp46, 24S-hydroxycholesterol, on the cholesterol regulatory genes ...

Could 24-S-hydroxycholesterol play a role in Müller glial cell's membrane dynamics in the rat

open access: yes, 2016
National audiencePurpose The catabolism of cholesterol in neurons leads to a more hydrophilic compound soluble form, the 24‐S‐hydroxycholesterol by means of an enzyme the CYP46A1.The aim of this study was to analyse the implication of 24‐S ...

Identification and in vivo detection of side-chain hydroxylated metabolites of 4β-hydroxycholesterol

open access: yes, 2023
Oxysterols are oxidized derivatives of cholesterol that are formed by enzymatic processes or through the action of reactive oxygen species. Several of these bioactive lipids have been shown to be affected and/or play a role in inflammatory processes.

Liver X Receptor α (LXRα) Regulates 5β‐Reductase (AKR1D1) Expression in Avian Embryos: Implications for Yolk Steroid Metabolism

open access: yesJournal of Experimental Zoology Part A: Ecological and Integrative Physiology, Volume 345, Issue 5, Page 456-461, June 2026.
ABSTRACT Steroid‐mediated maternal effects are well‐studied as a source of phenotypic variation. In bird eggs, the yolk contains various steroids that can influence embryonic development. However, one complicating factor in understanding how yolk steroids affect development is that the embryo metabolizes yolk steroids to regulate exposure.

Studies on the oxysterols 4alpha- and 4beta-hydroxycholesterol

open access: yes, 2011
Cytochrome P450 (CYP) enzymes catalyze the metabolism of both foreign and endogenous substances such as drugs and steroids. Enzymes in subfamily CYP3A sum up to 30% of the total CYP content in the liver and metabolize about 50% of all drugs.
