An adhesive hydrogel depot with polyphenol‐armored nanogenes is developed for treating aged osteoarthritis. This system maintains miR‐140 bioactivity, prolongs its intra‐articular retention, and enhances transfection into chondrocytes, leading to the downregulation of senescence‐associated pathways.

Local stress concentration disrupts metabolic homeostasis and induces inflammation in the nucleus pulposus (NP), thereby accelerating intervertebral disc degeneration (IDD). A biomimetic HA/ChS hydrogel millimeter sphere (ChS@HM) is developed to enable synergistic stress dispersion and sustained hydration lubrication.

Gel-like behavior in aggrecan assemblies [PDF]
Aggrecan, a large biological polyelectrolyte molecule with a bottlebrush shape, forms complexes with hyaluronic acid (HA) that provide compressive resistance in cartilage. In solutions of aggrecan alone, the concentration dependence of the osmotic pressure Π is marked by self-assembly of the molecules into aggregates.

Lesion Site‐Targeted Microspheres Modulate Nav1.7‐Related Signaling for Osteoarthritis Treatment
Cartilage‐targeted carbamazepine‐loaded WYRGRL‐modified composite microspheres (CBZ/WCOM) anchor to exposed type II collagen in osteoarthritic lesions and release carbamazepine under acidic conditions. This bind‐then‐release platform modulates Nav1.7‐related sodium signaling, Na⁺/Ca²⁺ exchanger‐associated Ca2+ dynamics, and heat shock protein 70 ...

The aim of this study was to find characteristic biomarkers in the serum of patients with osteoarthritis (OA) and psoriatic arthritis (PsA) responsible for inflammation and destruction of joint cartilage, which could differentiate these two diseases. The

The Glycosaminoglycan Attachment Regions of Human Aggrecan [PDF]
Aggrecan possesses both chondroitin sulfate (CS) and keratan sulfate (KS) chains attached to its core protein, which reside mainly in the central region of the molecule termed the glycosaminoglycan-attachment region. This region is further subdivided into the KS-rich domain and two adjacent CS-rich domains (CS1 and CS2).

Both SVF and CD31+ SVF improve spontaneous osteoarthritis, but CD31+ enrichment refines the therapeutic profile rather than broadly enhancing all effects. More distinct benefits are observed in extracellular matrix remodeling, osteochondral integrity, and subchondral/trabecular bone preservation, supporting CD31+‐enriched SVF as a biologically enhanced

THBS1+ Macrophages Exacerbate Modic Changes via SDC4‐Dependent Activation of NLRP3 Inflammasome
The illustration of THBS1+ macrophages exacerbate MCs via SDC4‐dependent activation of the NLRP3 inflammasome. THBS1+ macrophage subpopulation was identified in MCs through single‐cell RNA sequencing. C. acnes and its metabolites activate THBS1 expression in macrophages via the TLR signaling pathway.

The Different Roles of Aggrecan Interaction Domains [PDF]
The aggregating proteoglycans of the lectican family are important components of extracellular matrices. Aggrecan is the most well studied of these and is central to cartilage biomechanical properties and skeletal development. Key to its biological function is the fixed charge of the many glycosaminoglycan chains, that provide the basis for the ...
