Nanozyme‐Reinforced miR‐197‐3p Delivery Resets Metabolic and Senescence Pathways to Rejuvenate Osteoarthritic Cartilage

open access: yesAdvanced Science, EarlyView.
ABSTRACT Osteoarthritis (OA) is a progressive and disabling joint disease driven by oxidative stress, chondrocyte senescence and extracellular matrix (ECM) degradation, yet lacks effective disease‐modifying treatments. In this study, we identified miR‐197‐3p as a previously unrecognized, cartilage‐protective miRNA significantly downregulated in both ...

Aggrecan and COMP Improve Periosteal Chondrogenesis by Delaying Chondrocyte Hypertrophic Maturation

open access: yesFrontiers in Bioengineering and Biotechnology, 2020
The generation of cartilage from progenitor cells for the purpose of cartilage repair is often hampered by hypertrophic differentiation of the engineered cartilaginous tissue caused by endochondral ossification.

Biomimetic Gradient Hydrogels Regulate Osteochondral Regeneration Microenvironment Remodeling via Spatiotemporal Programming Engineering

open access: yesAdvanced Science, EarlyView.
Electric field‑driven continuous gradient hydrogel GHZF4 achieves spatiotemporal growth factor release, modulates immune microenvironment, angiogenesis and osteochondral differentiation. It realizes cell‑free functional osteochondral repair in rat/rabbit models, offering a promising biomimetic strategy for gradient tissue regeneration.

Catabolism of aggrecan, decorin and biglycan in tendon [PDF]

open access: yesBiochemical Journal, 2000
We have examined the catabolism of the proteoglycans aggrecan, decorin and biglycan in fresh tendon samples and in explant cultures of tissue from the tensional and compressed regions of young and mature bovine tendons. A panel of well-characterized antibodies that recognize glycosaminoglycan or protein (linear or neoepitope) sequences was used to ...

Kaempferol Attenuates Spaceflight‐Associated Knee Cartilage Degradation by Targeting NOX4‐Mediated Mitochondrial Dysfunction

open access: yesAdvanced Science, EarlyView.
We have shown that both simulated and actual spaceflight contribute to cartilage degradation in the knees of mice. Using RNA sequencing and bioenergetic profiling, we identified NADPH oxidase 4 (NOX4) as a key factor driving these changes. Additionally, we found that kaempferol, a naturally occurring flavonoid that binds directly to NOX4, reduces ...

Lateral Nanomechanics of Cartilage Aggrecan Macromolecules [PDF]

open access: yesBiophysical Journal, 2007
To explore the role of the brush-like proteoglycan, aggrecan, in the shear behavior of cartilage tissue, we measured the lateral resistance to deformation of a monolayer of chemically end-attached cartilage aggrecan on a microcontact printed surface in aqueous NaCl solutions via lateral force microscopy.

Deep Learning Prediction of O‐Glycopeptide Tandem Mass Spectra Enhances O‐Glycoproteomics

open access: yesAdvanced Science, EarlyView.
DeepGPO integrates Transformer and graph neural networks with tailored training strategies, including data augmentation, loss re‐weighting, and pre‐training, to achieve high‐quality O‐glycopeptide MS/MS spectra prediction. The predicted MS/MS spectra are used to localize O‐glycosylation sites from HCD MS/MS data, enhancing O‐glycoproteomics analysis ...
