Investigation of bioorganometallic artemisinins as antiplasmodials

The aim of this study was to improve the activity of artemisinin-based compounds for the treatment of malaria, primarily caused by Plasmodium falciparum. The artemisinin scaffold was functionalized with an appropriate N,N’– coordinated ruthenium p-cymene complex or ferrocene complex.

Chemical optimization of a novel pyridylpiperazine class yields two promising antimalarial candidates with an exceptional balance of metabolic stability and solubility. These optimized leads significantly protect mice from cerebral malaria, reducing parasitic load by up to 91.7%.

Promiscuous stimulation of HSP70 ATPase activity by parasite‐derived J‐domains

The malaria parasite Plasmodium falciparum exports three highly homologous yet functionally divergent J‐domain proteins into human erythrocytes. Here, we show that J‐domains isolated from all three proteins effectively stimulate the ATPase activity of both endogenous host and exported parasite HSP70 chaperones.
