Crosstalk between ATF4 and MTA1/HDAC1 promotes osteosarcoma progression
The stress response gene activating transcription factor 4 (ATF4) is involved in metastatic behavior and cellular protection. Here we show that ATF4 is upregulated in osteosarcoma (OS) cell lines and patient clinical samples as compared to matched non-tumor tissue.

Novel drugs approved by the EMA, the FDA and the MHRA in 2025: A year in review
Abstract In the 2025 novel drug mini‐review, one can take a full measure of the ingenuity that underlies current drug design and development, despite the year's smaller harvest (46 novel drugs) compared to 2024 (53) and 2023 (70). 54% of the novel drugs are first‐in‐class (FIC).

Inhibitors of insulin‐degrading enzyme boost PI cytotoxicity through an increased sensitivity of proteasome to PI inhibitors, induction of ISR, DNA damage and Myc down‐regulation. They overcome PI resistance in vitro and induce tumour regression in vivo.

Transcriptional landscape of neuronal and cancer stem cells [PDF]
Tumor mass is composed by heterogeneous cell population including a subset of “cancer stem cells” (CSC). Oncogenic signals foster CSC by transforming tissue stem cells or by reprogramming progenitor/differentiated cells towards stemness.

We designed a framework for screening clinical drug combinations with anti‐hepatocellular carcinoma (HCC) activity, comprising four parts: primary screening, single‐cell screening, functional validation, and mechanism research. High‐throughput single‐cell screening identifies HY (HHT and YM155) as a potent anti‐HCC drug combination, validated by in ...

ATF4 Regulates CD4+ T Cell Immune Responses through Metabolic Reprogramming
Summary: T cells are strongly regulated by oxidizing environments and amino acid restriction. How T cells reprogram metabolism to adapt to these extracellular stress situations is not well understood.

In the oxygen‐induced retinopathy (OIR) mouse model, two distinct microglial subpopulations play opposing roles: highly glycolytic microglia (HGM) promote neovascular formation via Pkm2, while Mrc1/CD206‐positive phagocytosis‐associated microglia (PAM) facilitate its regression.
