Dependence of Site-2 Protease Cleavage of ATF6 on Prior Site-1 Protease Digestion Is Determined by the Size of the Luminal Domain of ATF6 [PDF]
ATF6 is an endoplasmic reticulum (ER) membrane-anchored transcription factor activated by regulated intramembrane proteolysis in the ER stress response. The release of the cytosolic transcription factor domain of ATF6 requires the sequential processing by the Golgi site-1 and site-2 proteases (S1P and S2P).

Inhibition of KDELR2 in a small fraction of tumor cells generates sustainable immunogenic cell death conditions within the tumor microenvironment. These conditions promote the regression of tumors in a T cell independent manner. During regression, macrophages prime T cells that subsequently provide systemic protection against recurrence. The potency of

Cows with ketosis during the perinatal period often experience damage to the liver, a core metabolic organ, thereby exacerbating the disease's inherent vicious cycle. This study indicates that serum 3‐IAA concentrations were reduced in ketotic cows and show a significant negative correlation with hepatic TG content.

Novel Organelle‐Based Intracellular Immunity With Mechanistic and Therapeutic Implications
A conceptual framework illustrating how PAMPs/DAMPs initiate barrier, innate, adaptive, and intracellular immune responses, with organelle‐based intracellular immunity serving as a central integrator linking metabolism, inflammatory signaling, and therapeutic interventions to restore immune homeostasis.

Beneficial effects of neuronal ATF6 activation in permanent ischemic stroke
Objective: Brain ischemia leads to the accumulation of unfolded/misfolded proteins in the endoplasmic reticulum (ER) lumen and consequently, ER stress. To help cells restore ER function, a series of adaptive stress response pathways, collectively termed the unfolded protein response (UPR), are activated.

Smart Nanodelivery Systems for Immunometabolic Modulation in Osteoarthritis
Smart nanodelivery systems deliver therapeutic payloads across the cartilage matrix to ensure their sustained accumulation within chondrocytes at sites of injury. By targeting specific molecular targets and signaling pathways, these platforms modulate the pathological synovium‐cartilage axis to restore immunometabolic homeostasis and drive tissue ...
