Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Single-molecule visualization of conformational changes and substrate transport in the vitamin B12 ABC importer BtuCD-F

open access: yesNature Communications, 2017
The vitamin B12 importer BtuCD-F is a type II ATP-binding cassette transporter. Here the authors use single-molecule fluorescence techniques to analyse ATP hydrolysis and substrate transport in individual BtuCD-F complexes embedded in liposomes, and ...

Solid-state NMR investigations of the ATP binding cassette multidrug transporter LmrA [PDF]

open access: yes, 2006
The development of resistance to multiple drugs is a major problem in treatment of number of infectious diseases and cancer. The phenomenon of multidrug resistance (MDR) is based on the synergetic interplay of a number of mechanisms such as target ...

Identifying transcription factors controlling the basal expression of human MRP4 highlights a substantial role for Sp1

open access: yesFEBS Open Bio, EarlyView.
The MRP4 transporter exports several drugs and signaling molecules. Here, we identified key promoter elements regulating basal MRP4 expression. Using reporter assays, we defined a conserved region with essential Sp1 and contributory Ets sites, which controlled basal MRP4 expression.

The Binding Specificity of OppA Determines the Selectivity of the Oligopeptide ATP-binding Cassette Transporter [PDF]

open access: yes, 2004
The purification and functional reconstitution of a five-component oligopeptide ATP-binding cassette transporter with a remarkably wide substrate specificity are described.

Inward- and outward-facing X-ray crystal structures of homodimeric P-glycoprotein CmABCB1

open access: yesNature Communications, 2019
P-glycoprotein, an ATP-binding cassette (ABC) transporter, extrudes a large variety of xenobiotics from the cell which protects tissues from toxins.

Hypoxia-activated prodrug TH-302 decreased survival rate of canine lymphoma cells under hypoxic condition. [PDF]

open access: yesPLoS ONE, 2017
We tested the hypotheses that hypoxic stimulation enhances growth potentials of canine lymphoma cells by activating hypoxia-inducible factor 1α (HIF-1α), and that the hypoxia-activated prodrug (TH-302) inhibits growth potentials in the cells.

Fingerprint of the mitochondrial ABC transporter Mdl1p from Saccharomyces cerevisiae [PDF]

open access: yes, 2006
The multidrug resistance like protein 1 (Mdl1p) belongs to the class of ATP binding cassette (ABC) transporters which comprise a large family of membrane proteins utilising ATP hydrolysis to drive up-hill transport of a wide variety of solutes across ...

Functional Blood‐Brain Barrier Crossing by Biomimetic M13 Phage Vectors for Targeted Neuronal Delivery

open access: yesAdvanced Healthcare Materials, EarlyView.
This study investigates the M13 bacteriophage as a biomimetic nanovector capable of crossing in vitro models of the blood–brain barrier. By exploiting peculiar transcellular pathways, M13 avoids lysosomal degradation and preserves its structural integrity and functionality.
