2-Hydroxyisobutyrylation and Phosphorylation Crosstalk Guides Metastasis Prediction and Immunotherapy in Esophageal Squamous Cell Carcinoma. [PDF]

open access: yesMedComm (2020)
This study deciphers the functional crosstalk between 2‐hydroxyisobutyrylation (Khib) and phosphorylation in driving esophageal squamous cell carcinoma (ESCC) metastasis. Through multi‐omics and machine learning, we developed a plasma protein signature for noninvasive detection of lymph node metastasis.

Overcoming Photodynamic Therapy Resistance: GSH-Responsive AIE Photosensitizer-Hydroxycamptothecin Nanoliposomes for Extrahepatic Cholangiocarcinoma. [PDF]

open access: yesAdv Healthc Mater
A GSH‐responsive AIE nanoplatform (TSH NPs) with RGD‐mediated tumor targeting co‐delivers a photosensitizer (TPA) and HCPT for extrahepatic cholangiocarcinoma. The system synergistically enhances PDT and chemotherapy by suppressing HIF‐1α/VEGF‐driven hypoxic adaptation, effectively overcoming PDT resistance.

Cilengitide treatment of newly diagnosed glioblastoma patients does not alter patterns of progression [PDF]

open access: yes, 2014
The integrin antagonist cilengitide has been explored as an adjunct with anti-angiogenic properties to standard of care temozolomide chemoradiotherapy (TMZ/RT → TMZ) in newly diagnosed glioblastoma.

Integrin-αvβ3 as a Therapeutic Target in Glioblastoma: Back to the Future?

open access: yesPharmaceutics, 2022
Glioblastoma (GBM), the most common primary malignant brain tumor, is associated with a dismal prognosis. Standard therapies including maximal surgical resection, radiotherapy, and temozolomide chemotherapy remain poorly efficient.

In vitro and in vivo drug disposition of cilengitide in animals and human [PDF]

open access: yesPharmacology Research & Perspectives, 2016
AbstractCilengitide is very low permeable (1.0 nm/sec) stable cyclic pentapeptide containing an Arg‐Gly‐Asp motif responsible for selective binding to αvβ3 and αvβ5 integrins administered intravenously (i.v.). In vivo studies in the mouse and Cynomolgus monkeys showed the major component in plasma was unchanged drug (>85%).

Cyclic RGD Pentapeptide Cilengitide Enhances Efficacy of Gefitinib on TGF-β1-Induced Epithelial-to-Mesenchymal Transition and Invasion in Human Non-Small Cell Lung Cancer Cells

open access: yesFrontiers in Pharmacology, 2021
During non-small cell lung cancer (NSCLC) progression, transforming growth factor (TGF)-β mediated epithelial-to-mesenchymal transition (EMT) is an important process leading to high mortality and poor prognosis.

Cilengitide inhibits attachment and invasion of malignant pleural mesothelioma cells through antagonism of integrins αvβ3 and αvβ5.

open access: yesPLoS ONE, 2014
Malignant pleural mesothelioma (MPM) is an almost invariably fatal, asbestos-related malignancy arising from the mesothelial membrane lining the thoracic cavities.

TGFβ enhances platelet-breast-cancer-cell interaction and promotes platelet aggregation. [PDF]

open access: yesFEBS J
TGFβ stimulation promotes EMT in MCF7 cells, increasing their ability to bind, activate, and aggregate PLT. This effect involves the upregulation of adhesion molecules Integrin‐αv and Galectin‐3. Inhibiting Integrin‐αv or Galectin‐3, through Cilengitide and GB1103, reduces PLT adhesion to MCF7 and PLT aggregation, suggesting a mechanism linking TGFβ ...

Cilengitide – Exceptional pseudopolymorphism of a cyclic pentapeptide

open access: yesEuropean Journal of Pharmaceutical Sciences, 2015
Cilengitide (Cil) represents a cyclic pentapeptide, cyclo-(Arg-Gly-Asp-D-Phe-N-MeVal). Existence of an anhydrate form (A1) and a tetrahydrate form Cil1(H2O)4 has been observed. Surprisingly the anhydrate form proved to be more stable in aqueous environment compared to the tetrahydrate form.
