Diverse functions of deadenylases in DNA damage response and genomic integrity

WIREs RNA, 2020
AbstractDNA damage response (DDR) is a coordinated network of diverse cellular processes including the detection, signaling, and repair of DNA lesions, the adjustment of metabolic network and cell fate determination. To deal with the unavoidable DNA damage caused by either endogenous or exogenous stresses, the cells need to reshape the gene expression ...

Novel interaction between CCR4 and CAF1 in rice CCR4–NOT deadenylase complex

Plant Molecular Biology, 2016
Rice is an important crop in the world. However, little is known about rice mRNA deadenylation, which is an important regulation step of gene expression at the post-transcriptional level. The CCR4-NOT1 complex contains two key components, CCR4 and CAF1, which are the main cytoplasmic deadenylases in eukaryotic cells.

The CCR4-NOT deadenylase activity contributes to generation of induced pluripotent stem cells

Biochemical and Biophysical Research Communications, 2016
Somatic cells can be reprogrammed as induced pluripotent stem cells (iPSCs) by introduction of the transcription factors, OCT3/4, KLF4, SOX2, and c-MYC. The CCR4-NOT complex is the major deadenylase in eukaryotes. Its subunits Cnot1, Cnot2, and Cnot3 maintain pluripotency and self-renewal of mouse and human embryonic stem (ES) cells and contribute to ...

Analysis of the Ccr4-Not deadenylase complex: a biochemical and computational approach

2017
In eukaryotic cells, the degradation of the mRNA poly(A) tail (deadenylation) is a crucial step in the regulation of gene expression. The Ccr4-Not complex is the major deadenylase enzyme involved in the mRNA deadenylation. The complex is composed by two subunits with ribonuclease activity (Caf1 and Ccr4) and at least six non-catalytic subunits.
