Acyl-protein thioesterase1 alleviates senile osteoporosis by promoting osteoblast differentiation via depalmitoylation of BMPR1a

open access: yesRegenerative Therapy, 2023
Objective: Senile osteoporosis (SOP) is an aging-related disease. The depalmitoylating enzyme Acyl-protein thiesterase1 (APT1) is involved in disease regulation. This study explored the mechanism of APT1 in SOP.

Greasing the wheels or a spanner in the works?:Regulation of the cardiac sodium pump by palmitoylation [PDF]

open access: yes, 2018
The ubiquitous sodium/potassium ATPase (Na pump) is the most abundant primary active transporter at the cell surface of multiple cell types, including ventricular myocytes in the heart.

Specific oligomerization of the 5-HT1A receptor in the plasma membrane [PDF]

open access: yes, 2009
In the present study we analyze the oligomerization of the 5-HT1A receptor within living cells at the sub-cellular level. Using a 2-excitation Foerster Resonance Energy Transfer (FRET) method combined with spectral microscopy we are able to estimate the ...

Protein Palmitoylation Modification During Viral Infection and Detection Methods of Palmitoylated Proteins

open access: yesFrontiers in Cellular and Infection Microbiology, 2022
Protein palmitoylation—a lipid modification in which one or more cysteine thiols on a substrate protein are modified to form a thioester with a palmitoyl group—is a significant post-translational biological process. This process regulates the trafficking,

Protein Depalmitoylation Is Induced by Wnt5a and Promotes Polarized Cell Behavior [PDF]

open access: yesJournal of Biological Chemistry, 2015
Wnt5a signaling regulates polarized cell behavior, but the downstream signaling events that promote cell polarity are not well understood. Our results show that Wnt5a promotes depalmitoylation of the melanoma cell adhesion molecule (MCAM) at cysteine 590.

Attenuated palmitoylation of serotonin receptor 5-HT1A affects receptor function and contributes to depression-like behaviors [PDF]

open access: yes, 2019
The serotonergic system and in particular serotonin 1A receptor (5-HT1AR) are implicated in major depressive disorder (MDD). Here we demonstrated that 5-HT1AR is palmitoylated in human and rodent brains, and identified ZDHHC21 as a major palmitoyl ...

Peroxiredoxin 6 mediates Gαi protein-coupled receptor inactivation by cJun kinase

open access: yesNature Communications, 2017
Opioid receptors are important modulators of nociceptive pain. Here the authors show that opioid receptor activation recruits peroxiredoxin 6 (PRDX6) to the receptor-Gαi complex by c-Jun N-terminal kinase, resulting in Gαi depalmitoylation and enhanced ...

Palmitoylation of Plasmodium alveolins promotes cytoskeletal function. [PDF]

open access: yes, 2017
S-palmitoylation is a post-translational lipid modification that is widespread among Plasmodium proteins and essential for parasite development. Little is known about the contribution of palmitoylation to the function of individual parasite molecules and

PIK3CA Mutations Downregulate PPT1 to Promote Adipogenesis by Suppressing P300 Depalmitoylation and Phase Separation

open access: yesAdvanced Science, EarlyView.
This study demonstrates that somatic PIK3CA mutations suppress PPT1 expression via activation of the PI3K–AKT–c‐JUN axis. This reduction in PPT1 weakens its interaction with P300, thereby increasing palmitoylation at C1176 of P300 and protecting P300 from lysosomal degradation.

Protein palmitoylation in cancer: molecular functions and therapeutic potential

open access: yesMolecular Oncology, 2023
Protein S‐palmitoylation (hereinafter referred to as protein palmitoylation) is a reversible lipid posttranslational modification catalyzed by the zinc finger DHHC‐type containing (ZDHHC) protein family.
