Inducing Pyroptosis via Gasdermin B and Gasdermin E Cleavage

2023
Gasdermin B (GSDMB) and gasdermin E (GSDME) are two members of the gasdermin family, which shares a conservative gasdermin-N domain capable of executing pyroptotic cell death, through perforating the plasma membrane from inside of the cell. Both GSDMB and GSDME are autoinhibited in the resting stage and require proteolytic cleavage to unleash the pore ...

Caspase‐3 and gasdermin E mediate macrophage pyroptosis in periodontitis

Journal of Periodontal Research, 2023
AbstractBackground and ObjectivesPeriodontitis is a chronic inflammatory disease linked to pyroptosis, an inflammatory cell death process. Macrophages are essential for maintaining microenvironment homeostasis, which is crucial for periodontal health.

Molecular mechanisms and therapeutic relevance of gasdermin E in human diseases

Cellular Signalling, 2022
Gasdermin E (GSDME) is one of the main members of the GSDM family and is originally involved in hereditary hearing loss. Recent studies have reported that GSDME expression is epigenetically silenced by methylation in several common tumours, thereby enhancing tumour proliferation and metastasis.

Role of Gasdermin E in the Biogenesis of Apoptotic Cell–Derived Exosomes

The Journal of Immunology, 2023
Abstract The gasdermins are a family of pore-forming proteins that has recently been suggested to play a central role in pyroptosis. In this study, we describe the novel roles of gasdermins in the biogenesis of apoptotic cell–derived exosomes.

Molecular characterization of a fungal gasdermin-like protein [PDF]

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2020
Programmed cell death (PCD) in filamentous fungi prevents cytoplasmic mixing following fusion between conspecific genetically distinct individuals (allorecognition) and serves as a defense mechanism against mycoparasitism, genome exploitation, and ...

The Gasdermin‐D pore acts as a conduit for IL‐1β secretion in mice

open access: yesEuropean Journal of Immunology, 2018
The pro-inflammatory cytokine IL-1β is well known for its role in host defense and the initiation of potent inflammatory responses. It is processed from its inactive pro-form by the inflammatory caspase-1 into its mature bioactive form, which is then ...

Zebrafish gasdermin E cleavage-engaged pyroptosis by inflammatory and apoptotic caspases

Developmental & Comparative Immunology, 2021
As the executor of pyroptosis known to date, gasdermins (GSDMs), consists of GSDMA, GSDMB, GSDMC, GSDMD, GSDME and pejvakin, might play critical roles in anti-bacterial infection as well as inflammatory diseases. However, zebrafish only harbors a pair of Gsdme (Gsdmea/b), and their activation mechanisms remain largely unknown.

Gasdermin-E mediates mitochondrial damage in axons and neurodegeneration

2022
Summary Mitochondrial dysfunction and axon loss are hallmarks of neurologic diseases. Gasdermin (GSDM) proteins are executioner pore-forming molecules that mediate cell death, yet their roles in the central nervous system (CNS) are not well understood.

MACC1 and Gasdermin-E (GSDME) regulate the resistance of colorectal cancer cells to irinotecan

Biochemical and Biophysical Research Communications, 2023
Metastasis-associated in colon cancer 1 (MACC1) is an oncogene associated with the progression and metastasis of many solid cancer entities. High expression of MACC1 is found in colorectal cancer (CRC) tissues. So far, the role of MACC1 in CRC cell pyroptosis and resistance to irinotecan is unclear.

Enhanced Gasdermin-E-mediated Pyroptosis in Alzheimer’s Disease

Neuroscience
Amyloid β protein (Aβ) is a critical factor in the pathogenesis of Alzheimer's disease (AD). Aβ induces apoptosis, and gasdermin-E (GSDME) expression can switch apoptosis to pyroptosis. In this study, we demonstrated that GSDME was highly expressed in the hippocampus of APP23/PS45 mouse models compared to that in age-matched wild-type mice.
