Gephyrin: where do we stand, where do we go?

open access: yes, 2008
Gephyrin is a multifunctional protein responsible for molybdenum cofactor synthesis and the clustering of glycine and GABA(A) receptors at inhibitory synapses.

Gephyrin: a master regulator of neuronal function?

open access: yesNature Reviews Neuroscience, 2014
The neurotransmitters GABA and glycine mediate fast synaptic inhibition by activating ligand-gated chloride channels--namely, type A GABA (GABA(A)) and glycine receptors. Both types of receptors are anchored postsynaptically by gephyrin, which self-assembles into a scaffold and interacts with the cytoskeleton.

The gephyrin scaffold modulates cortical layer 2/3 pyramidal neuron responsiveness to single whisker stimulation

open access: yesScientific Reports
Gephyrin is the main scaffolding protein at inhibitory postsynaptic sites, and its clusters are the signaling hubs where several molecular pathways converge.

Strukturelle und biochemische Charakterisierung von Gephyrin und verschiedenen Gephyrin-Liganden-Komplexen

open access: yes, 2014
Efficient synaptic neurotransmission requires the exact apposition of presynaptic terminals and matching neurotransmitter receptor clusters on the postsynaptic side.

Neurexin-1β Binding to Neuroligin-1 Triggers the Preferential Recruitment of PSD-95 versus Gephyrin through Tyrosine Phosphorylation of Neuroligin-1

open access: yesCell Reports, 2013
Adhesion between neurexin-1β (Nrx1β) and neuroligin-1 (Nlg1) induces early recruitment of the postsynaptic density protein 95 (PSD-95) scaffold; however, the associated signaling mechanisms are unknown.

The E3 Ubiquitin Ligase UBE3B Regulates Synaptic Development and Cortical Network Activity

open access: yesAutism Research, Volume 19, Issue 5, May 2026.
ABSTRACT Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by impaired communication, abnormal social interactions, and restricted, repetitive behaviors. Pathogenic mutations in UBE3B result in neurodevelopmental disease, including intellectual disability, lack of speech, and ASD.

Enrichment of GABAA receptor α-subunits on the axonal initial segment shows regional differences

open access: yesFrontiers in Cellular Neuroscience, 2016
Although it is generally recognized that certain α-subunits of γ-aminobutyric acid type A receptors (GABAARs) form enriched clusters on the axonal initial segment (AIS), the degree to which these clusters vary in different brain areas is not well known ...

Gephyrin is palmitoylated by DHHC-12.

open access: yes, 2014
(a) Individual HA-tagged DHHC proteins or GST-HA as control were co-expressed with gephyrin-GFP for 24 h in HEK293 cells and analyzed with the ABE assay. Omitting hydroxylamine demonstrated specify of the assay.

Distinct roles for extracellular and intracellular domains in neuroligin function at inhibitory synapses

open access: yeseLife, 2016
Neuroligins (NLGNs) are postsynaptic cell adhesion molecules that interact trans-synaptically with neurexins to mediate synapse development and function. NLGN2 is only at inhibitory synapses while NLGN3 is at both excitatory and inhibitory synapses.
