Atypical Iron-Sulfur Cluster Binding, Redox Activity and Structural Properties of Chlamydomonas reinhardtii Glutaredoxin 2

open access: yesAntioxidants, 2021
Glutaredoxins (GRXs) are thioredoxin superfamily members exhibiting thiol-disulfide oxidoreductase activity and/or iron-sulfur (Fe-S) cluster binding capacities. These properties are determined by specific structural factors.

Early oxidation‐related changes in aqueous humor following eye exposure to ultraviolet‐A radiation

open access: yesPhotochemistry and Photobiology, EarlyView.
UVA exposure induces early oxidative alterations in the aqueous humor, disrupting its antioxidant balance. Levels of ascorbic acid, reduced glutathione (GSH), and the GSH/GSSG ratio decrease, while oxidized glutathione (GSSG), S‐glutathionylated proteins (PSSG), and malondialdehyde (MDA), a lipid peroxidation by‐product, increase.

Glutathione and Glutaredoxin—Key Players in Cellular Redox Homeostasis and Signaling

open access: yesAntioxidants, 2023
This Special Issue of Antioxidants on Glutathione (GSH) and Glutaredoxin (Grx) was designed to collect review articles and original research studies focused on advancing the current understanding of the roles of the GSH/Grx system in cellular homeostasis

Ageing alters cysteine oxidation‐regulated redox signalling in skeletal muscle: Integrative omics and AI‐based structural predictions

open access: yesExperimental Physiology, EarlyView.
Abstract Ageing is associated with loss of skeletal muscle mass and strength (sarcopenia) and disrupted redox homeostasis. Redox signalling is essential for muscle adaptation, yet the mechanisms by which ageing disrupts cysteine‐based regulation are poorly defined. The drivers of site‐specific reactivity and signalling specificity in aged muscle remain

Canonical and non‐canonical functions of proteins regulating mitochondrial dynamics in mammalian physiology

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend Mitochondria are highly dynamic organelles that continuously remodel their architecture through coordinated cycles of fusion and fission. This review examines the four key GTPases that orchestrate mitochondrial dynamics in mammals: MFN1, MFN2, OPA1, and DRP1.

Dicarbonyl Stress and S-Glutathionylation in Cerebrovascular Diseases: A Focus on Cerebral Cavernous Malformations

open access: yesAntioxidants, 2020
Dicarbonyl stress is a dysfunctional state consisting in the abnormal accumulation of reactive α-oxaldehydes leading to increased protein modification.

Mitochondrial Ca2+ uniporter haploinsufficiency leads to sexually dimorphic redox imbalance and metabolic remodelling in the mouse brain

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend The genetic inactivation of one Mcu allele leads to sex‐specific changes in neuronal function in adult mice, that is, the firing of action potentials and the relationship between cytosolic and mitochondrial Ca2+ levels. The ability to produce NAD(P)H by stimulated neural tissue is largely preserved in male mice but delayed in ...

Identification of DUOX1-dependent redox signaling through protein S-glutathionylation in airway epithelial cells

open access: yesRedox Biology, 2014
The NADPH oxidase homolog dual oxidase 1 (DUOX1) plays an important role in innate airway epithelial responses to infection or injury, but the precise molecular mechanisms are incompletely understood and the cellular redox-sensitive targets for DUOX1 ...

Deciphering pro‐arrhythmogenic mechanisms of EPAC in human atrial cardiomyocytes

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend This study aimed to investigate the effect of exchange proteins directly activated by cAMP (EPAC) on the regulation of human atrial cardiomyocyte electrophysiology and their potential involvement in the onset of atrial fibrillation (AF).

The busulfan metabolite EdAG irreversibly glutathionylates glutaredoxins [PDF]

open access: yesArchives of Biochemistry and Biophysics, 2015
The DNA alkylating agent busulfan is used to 'precondition' patients with leukemia, lymphomas and other hematological disorders prior to hematopoietic stem cell transplants. Busulfan is metabolized via conjugation with glutathione (GSH) followed by intramolecular rearrangement to the GSH analog γ-glutamyl-dehydroalanyl -glycine (EdAG).
