Inhibition of Insulin-Regulated Aminopeptidase: A Novel Therapeutic Strategy for Stroke

open access: yes, 2022
This thesis characterises and validates a preclinical model of stroke that closely mimics human stroke. In this preclinical model, novel drug therapies that target the insulin-regulated aminopeptidase (IRAP), an enzyme found in the brain, were tested ...

IRAP Inhibitors: M1-Aminopeptidase Family Inspiration

open access: yesFrontiers in Pharmacology, 2020
The insulin regulated aminopeptidase (IRAP) has been proposed as an important therapeutic target for indications including Alzheimer’s disease and immune disorders.

Insulin-Regulated Aminopeptidase in Women with Breast Cancer: A Role beyond the Regulation of Oxytocin and Vasopressin. [PDF]

open access: yesCancers (Basel), 2020
Insulin-regulated aminopeptidase (IRAP) is the only enzyme known to cleave oxytocin and vasopressin; however, it is also the high-affinity binding site for angiotensin IV (AngIV) receptor type 4 (AT4) ligands and it is related to insulin-dependent glucose transporters through the translocation of the glucose transporter type 4 (GLUT4). Previous studies

Hydrogen Peroxide Induced Toxicity Is Reversed by the Macrocyclic IRAP-Inhibitor HA08 in Primary Hippocampal Cell Cultures

open access: yesCurrent Issues in Molecular Biology, 2022
Angiotensin IV (Ang IV), a metabolite of Angiotensin II, is a bioactive hexapeptide that inhibits the insulin-regulated aminopeptidase (IRAP). This transmembrane zinc metallopeptidase with many biological functions has in recent years emerged as a new ...

Pleiotropic effects of Syntaxin16 identified by gene editing in cultured adipocytes

open access: yesFrontiers in Cell and Developmental Biology, 2022
Adipocytes play multiple roles in the regulation of glucose metabolism which rely on the regulation of membrane traffic. These include secretion of adipokines and serving as an energy store.

Translocation of the insulin-regulated aminopeptidase to the cell surface: detection by radioligand binding. [PDF]

open access: yesBr J Pharmacol, 2008
Background and purpose:Insulin‐regulated aminopeptidase (IRAP) and the insulin‐dependent glucose transporter GLUT4 colocalize in specific intracellular vesicles (that is, GLUT4 vesicles). These vesicles move slowly to the cell surface, but their translocation is markedly enhanced by insulin, resulting in higher glucose uptake.

Crystal structure of human insulin-regulated aminopeptidase with specificity for cyclic peptides. [PDF]

open access: yesProtein Sci, 2015
AbstractInsulin‐regulated aminopeptidase (IRAP or oxytocinase) is a membrane‐bound zinc‐metallopeptidase that cleaves neuroactive peptides in the brain and produces memory enhancing effects when inhibited. We have determined the crystal structure of human IRAP revealing a closed, four domain arrangement with a large, mostly buried cavity abutting the ...

Characterization of the Insulin-regulated Membrane Aminopeptidase in 3T3-L1 Adipocytes [PDF]

open access: yesJournal of Biological Chemistry, 1996
A novel membrane aminopeptidase has been identified as a major protein in vesicles from rat adipocytes containing the glucose transporter isotype Glut4. In this study we have characterized this aminopeptidase, referred to as vp165, in 3T3-L1 adipocytes. The subcellular distributions of vp165 and Glut4 were determined by immunoisolation of vesicles with

The Type of Fat in the Diet Influences the Behavior and the Relationship Between Cystinyl and Alanyl Aminopeptidase Activities in Frontal Cortex, Liver, and Plasma

open access: yesFrontiers in Molecular Biosciences, 2020
Insulin-regulated aminopeptidase (IRAP, cystinyl aminopeptidase, CysAP) and aminopeptidase M (alanyl aminopeptidase, AlaAP) are closely related enzymes involved in cognitive, metabolic, and cardiovascular functions.

Local renin angiotensin system and sperm DNA fragmentation

open access: yesAsian Journal of Andrology, 2022
The renin angiotensin system (RAS) appears to influence male fertility at multiple levels. In this work, we analyzed the relationship between the RAS and DNA integrity.
