Stabilization of the open conformation οf insulin-regulated aminopeptidase by a novel substrate-selective small-molecule inhibitor. [PDF]

open access: yesProtein Sci
Insulin-regulated aminopeptidase (IRAP) is an enzyme with important biological functions and the target of drug-discovery efforts. We combined in silico screening with a medicinal chemistry optimization campaign to discover a nanomolar inhibitor of IRAP ...

Insulin-regulated aminopeptidase in adipocyte is Cys-specific and affected by obesity [PDF]

open access: yesJournal of Molecular Endocrinology, 2015
Insulin-regulated aminopeptidase (IRAP, EC 3.4.11.3) in adipocytes is well known to traffic between high (HDM) and low (LDM) density microsomal fractions toward the plasma membrane (MF) under stimulation by insulin. However, its catalytic preference for aminoacyl substrates with N-terminal Leu or Cys is controversial.

Proteomic Analysis of Regulated Dendritic Cell Endosomes Reveals Dynamic Adaptation to Antigen Uptake and Cross-Presentation. [PDF]

open access: yesEur J Immunol
Phagocytosis of yeast activates recruitment of proteases and trafficking factors from the secretory and endosomal pathways to the IRAP compartment and subsequently phagosomes, promoting cross‐presentation. In the absence of Sec22b, IRAP endosome and phagosome maturation to late endosomes is accelerated.

Intrathecal injections of non-peptide oxytocin receptor agonists, WAY 267,464 and TC OT 39, induce significant anti-hyperalgesia in both male and female rats with inflammatory pain [PDF]

open access: yesIranian Journal of Basic Medical Sciences
Objective(s): Hyperalgesia is a clinical condition related to chronic pain in which patients experience increased nociceptive sensitivity. Intrathecal oxytocin has been shown to induce significant anti-hyperalgesia in both rodents and humans. However, in

Insulin and glucose play a role in foam cell formation and function

open access: yesCardiovascular Diabetology, 2006
Background Foam cell formation in diabetic patients often occurs in the presence of high insulin and glucose levels. To test whether hyperinsulinemic hyperglycemic conditions affect foam cell differentiation, we examined gene expression, cytokine ...

Anti-Inflammatory and Antitumor Effects of Hydroxytyrosol but Not Oleuropein on Experimental Glioma In Vivo. A Putative Role for the Renin-Angiotensin System

open access: yesBiomedicines, 2018
Functional roles of the angiotensin peptides of the renin-angiotensin system (RAS) cascade can be analyzed through their corresponding proteolytic regulatory enzymes aspartyl aminopeptidase (ASAP), aminopeptidase A (APA), aminopeptidase B (APB ...

Crystal structure of insulin-regulated aminopeptidase with bound substrate analogue provides insight on antigenic epitope precursor recognition and processing

open access: yes, 2015
Aminopeptidases that generate antigenic peptides influence immunodominance and adaptive cytotoxic immune responses. The mechanisms that allow these enzymes to efficiently process a vast number of different long peptide substrates are poorly understood ...

The emerging multifunctional roles of ERAP1, ERAP2 and IRAP between antigen processing and renin-angiotensin system modulation

open access: yesFrontiers in Immunology, 2022
The Endoplasmic Reticulum Aminopeptidase 1 and 2 (ERAP1 and ERAP2) and Insulin Regulated Aminopeptidase (IRAP) are three M1 zinc metalloproteases whose role in antigen processing is the refining of peptidome either in the Endoplasmic reticulum (ERAP1 and

Serum IRAP, a Novel Direct Biomarker of Prediabetes and Type 2 Diabetes?

open access: yesFrontiers in Molecular Biosciences, 2021
Insulin resistance (IR), currently called prediabetes (PD), affects more than half of the adult population worldwide. Type 2 diabetes (T2D), which often follows in the absence of treatment, affects more than 475 million people and represents 10–20% of ...

From biology to biotechnology: Host‐regulation factors from parasitoid wasps are a source of bioactive molecules with translational potential

open access: yesInsect Molecular Biology, EarlyView.
Parasitoid wasps deploy maternal and embryonic factors to reprogramme host physiology. Venom, calyx fluid, polydnaviruses, teratocytes and larval secretions act in a coordinated, compartmentalised manner. Host‐regulation factors are promising sources of insecticidal, antimicrobial and bioinspired translational molecules.
