Abstract Protein aggregation is increasingly recognized as a biologically relevant process in viral proteins, yet the molecular determinants governing such phenomena remain poorly understood. Intrinsically disordered proteins (IDPs) are ubiquitous in viral proteomes, where conformational plasticity not only enables functional diversity but also permits

ABSTRACT The role of the cell envelope‐associated Rv0132c/FGD2 from Mycobacterium tuberculosis has long been a subject of debate. Importantly, FGD2 is found only in pathogenic mycobacteria, making it a potential drug target. While some suggest it functions as a glucose‐6‐phosphate dehydrogenase, others propose it acts instead as an F420‐dependent ...

Abstract Background and Purpose β2‐adrenoceptor (β2AR) agonists are the cornerstone of asthma therapy and promote bronchodilation through Gαs signalling in airway smooth muscle (ASM), but their efficacy is limited by β‐arrestin‐mediated β2AR desensitization.

We present a set of multifunctional tacrine derivatives, designed with the CADMA‐Chem protocol, aimed for Alzheimer and Parkinson diseases. Of the tacrines designed, TAC‐674 and TAC‐678 are the best candidates to act as multifunctional antioxidants and neuroprotectors for Alzheimer and Parkinson diseases, respectively.

Mapping Trofinetide Polypharmacology in Rett Syndrome: A Multi‐Stage Computational Analysis
How can a single molecule address the complex symptoms of Rett syndrome? This computational study maps the multi‐target mechanism of trofinetide, revealing diverse modes of binding across five key receptors: GAT1, GABAA, CHRM1, AMPA, and GSK3β. From deep orthosteric anchoring to dynamic surface interactions, these structural insights provide a rational

Interpreting the effects of DNA polymerase variants at the structural level
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...
